Objective: To investigate the effect of PRP on the regeneration of hair bulb in the upper segment of hair follicles of rats' tentacles.Methods:(1)10 SD male rats were selected,and improved micro-separation technique was adopted to isolate the hair follicles at the growing stage after satisfactory anesthesia.The exposed red hairballs in the middle segment of the hair follicles were squeezed with tweezers,and 150 tissues in the upper segment of the hair follicles were obtained by transversely cutting close to the upper of the red hairballs.(2)Extract rats' PRP with twice centrifugation,and obtain PRP supernatant after activation;add different concentrations of PRP supernatant into culture media,preparing three kinds of culture medium: 0,5% and 10% PRP,and 90 tissues were cultured in the three medium respectively and in the same incubator of 37 ? and 5% CO2 for 16 days in vitro,every 48 h renewing the medium.(3)60 tissues from the upper segment of hair follicles were cultured in vitro in the three culture medium for 1 days,and then transplanted to the back skin of nude mice respectively.After 4 weeks,nude mice were killed and harvested.(4)Observe the tissue growth of the upper segment of hair follicles cultured in vitro with optical microscope,ALP of tissues cultured in vitro was validated by histology detection,regeneration of hair bulb tissues of upper segment of hair follicles was observed by HE staining,and immunohistochemical was applied to observe ?-SMA expression of new tissues of hair follicles.The number of hair follicles regenerated both in vitro and in vivo was calculated.Results:(1)PRP culture media could boost the regeneration of the hair bulb in hair follicles both in vivo and in vitro with similar structure of hair ball formed in vitro and visibly complete and mature hair ball regenerated inside nude mice.(2)5% PRP can significantly improve the activity of ALP and boost the regeneration rate in the upper segment of hair follicles in vitro.(3)?-SMA was positive in the new tissue cultured both in vivo and in vitro.Conclusion: PRP can promote the regeneration of the upper segment of hair follicle,and 5%PRP is better than 10%PRP in vitro experiments.
