Objective:To study the effect of Polygonum cuspidatum extract Resveratrol(Res)on the growth of human lung adenocarcinoma A549 cells and the expression of Nuclear Factor-?B(NF-?B)P65.Method:Take human lung adenocarcinoma A549 cells cultured in vitro,add0umol/L,30umol/L,50umol/L,100umol/L resveratrol intervention for 24 hours and 48 hours,determination of proliferation of A549 cells using Cell Counting Kit-8(CCK-8).Reverse Transcription-Polymerase Chain Reaction(RT-PCR)was used to detect the effect of resveratrol on the expression of NF-?B P65 m RNA in A549 cells,analysis of cell cycle and apoptosis of A549 cells by flow cytometry,detection of NF-?B P65 protein expression in A549 cells by Western blot.Result:The CCK-8 test results showed that resveratrol could inhibit the proliferation of human lung adenocarcinoma A549 cells,and the inhibitory effect became stronger with increasing concentration and time.After the human lung adenocarcinoma A549 cells were treated with the above-mentioned different concentrations of resveratrol for 48 hours,RT-PCR results showed that resveratrol could down-regulate the expression of NF-?B P65 m RNA,and with the increase of the concentration of resveratrol(30umol/L,50umol/L,100umol/L),the lower the relative expression of NF-?B P65 m RNA.By Western blot,we found that the expression of NF-?B P65 protein was significantly reduced in human lung adenocarcinoma A549 cells treated with different concentrations of resveratrol.After 48 hours of resveratrol action on human lung adenocarcinoma A549 cells,flow cytometry showed that as the concentration of resveratrol increased,the percentage of G0-G1 phase in human lung adenocarcinoma A549 cells significantly increased,reduced the percentage of S phase,blocked the cell cycle,and promoted the early and late apoptotic rates of human lung adenocarcinoma A549 cells.Conclusion:1.Resveratrol can reduce the proliferation of human lung adenocarcinoma A549 cells by blocking the cell cycle and inducing apoptosis,while down-regulating the expression of NF-?B P65 in human lung adenocarcinoma A549 cells,2.Resveratrol may inhibit lung adenocarcinoma progression by down-regulating NF-?B P65.
