Objective: To study the influence of the IL-6 and resveratrol on A549 cell damage induced by LPS and its intervention on proliferating of the A549 cells. To study the mechanism of A549 cell apoptosis induced by resveratrol and construct the SiRNA of IL-6 in A549 cell line.Method: In this study, cultured A549 cells was processed by adding IL-6,resveratrol and LPS in advance. Proliferation effect of A549 cells and influences of different drugs on it were examined by MTT ; A549 cell apoptosis induced by resveratrol was studied by TUNEL stain, and changes of the apoptosis-related proteins expression were detected by Western blotting. Sets of IL-6 siRNA oligo were synthesized by chemosynthesis, which were transfected through liposome; The expression of IL-6 and its subsequent changes after LPS stimulation was examined by RT-PCR.Result:1. LPS could damage cultured A549 cells in vitro, which is time and dosedependent. Extrinsic IL-6 has no significant intervention effect on A549cells damage induced by LPS. 2. Resveratrol has no significant intervention effect on A549 cells damage induced by LPS, while resveratrol has obvious inhibitory action on the proliferation of A549 cells, and the effect was mediated by apoptosis , which was carried out by inhibiting the expression and phosphorylation of Akt and modulating the expression of the apoptosis-related proteins, while it didn't influence the expression of related proteins by death receptor pathway.3. IL-6 expression exists in cultured A549 cells. The expression level was varied with LPS stimulation and interference of IL-6 SiRNA transfected by liposome.Conclusion: A549 cells damage induced by LPS was not affected by extrinsic IL-6,resveratrol in this study. Resveratrol has obvious inhibitory action on the proliferation of A549 cells and could induce A549 cells apoptosis by modulating the exprssion of apoptosis-related proteins. IL-6 expression level detected in cultured A549 cells was proportional to the amount of LPS induction. IL -6 SiRNA was constucted successfully.
