Objective:To compare the degree of fibrosis in eutopic endometrium and ectopic lesions and explore the effect of abdominal microenvironment on fibrosis by establishing a mouse model of endometriosis.Wenshen Xiaozheng Decoction was administered to the stomach.Comparing the degree of ectopic fibrosis in each group,and exploring the effect of Wenshen Xiaozheng Decoction on endometriosis fibrosis through IL-33 by Wenshen Xiaozheng Decoction and IL-33 intervention model mice;By observing the expression changes of IL-33 in the peritoneum,omentum,and mesentery of EMs mice under the intervention of Wenshen Xiaozheng Decoction,the possible sites of downregulation of IL-33 by Wenshen Xiaozheng Decoction were investigated.Methods:Part ?1.Comb the basis of previous research.IL-33 can promote fibrosis of endometriosis;Wenshen Xiaozheng Decoction may improve the fibrosis of endometriosis by down-regulating the expression of IL-33 in endometriosis serum,peritoneal fluid and ectopic lesions Into.Part ?1.Establish a mouse model of endometriosis by intraperitoneal injection.2.The model mice were randomly divided into a model group,a Wenshen Xiaozheng Decoction group,a Wenshen Xiaozheng Decoction+ rIL-33 group.Each group of mice was orally administered for 21 consecutive days.(1)Wenshen Xiaozheng Decoction group:mice were administered 0.2ml of Wenshen Xiaozheng Decoction daily at a dose of 11.678 g/kg,and saline was injected intraperitoneally twice a week.(2)Wenshen Xiaozheng Decoction+rHL-33 group:0.2ml Wenshen Xiaozheng Decoction was administered to mice at a daily dose of 11.678 g/kg,and 0.1ml rIL-33 was intraperitoneally injected twice a week at a dose of 0.05 ?g/g/d.(3)Model group:0.2ml of sterile distilled water is administered daily,and saline is injected intraperitoneally twice a week.3.The eutopic endometrium and ectopic lesions of the model group were taken,the collagen fiber area was detected by Masson staining,and the expressions of a-SMA and Collagen I were detected by immunohistochemistry.4.The model group,Wenshen Xiaozheng Decoction group,Wenshen Xiaozheng Decoction+rIL-33 group were used to detect ectopic lesions,Masson staining was used to detect collagen fiber area,and immunohistochemical method was used to detect the expression of a-SMA and Collagen I.5.The peritoneal tissue,omentum,and mesentery of the model group and Wenshen Xiaozheng Decoction group were taken,and the expression of IL-33 was detected by immunohistochemistry.Results:1.The collagen fiber area ratio,?-SMA,and Collagen I expression of ectopic lesions in the model group were higher than those in the eutopic endometrium(P<0.05).2.Compared with the ectopic lesions in the model group,the collagen fiber area ratio,a-SMA,and Collagen I expression of the ectopic lesions in the Wenshen Xiaozheng Decoction group decreased(P<0.05),and the ectopic lesions in the Wenshen Xiaozheng Decoction+rIL-33 group Collagen fiber area ratio,?-SMA,and Collagen I expression increased(P<0.05).3.In endometriosis model mice,the expression of IL-33 was higher in the omentum and mesentery,and the expression of IL-33 in peritoneum was less(P<0.05).Compared with the model group,the expression of IL-33 in the mesentery decreased in the Wenshen Xiaozheng Decoction group(P<0.05),and there was no significant difference between the omentum and mesentery(P>0.05).Conclusion:1.The degree of fibrosis in ectopic lesions of endometriosis is higher than that of the endometrium,suggesting that the abdominal microenvironment may promote the formation of fibrosis.2.Wenshen Xiaozheng Decoction may down-regulate IL-33,inhibit the formation of ectopic fibrosis,and improve the degree of endometriosis fibrosis,suggesting that Wenshen Xiaozheng Decoction may improve the endometriosis fibrosis.3.Wenshen Xiaozheng Decoction may down-regulate the expression of IL-33 in the mesentery and improve endometriosis fibrosis.
