Objective: To investigate the therapeutic effect of Huaihua San on ulcerative colitis model rats and its mechanism,and to provide basic experimental basis for the clinical application of Huaihua San.Methods: Using the method of animal experiment,32 SD male rats were divided into 4 groups according to the average of 8 rats in each group by using the random number table method,which were labeled as blank control group,model group,Huaihua San group and mesalazine group.Rats do not deal with blank control group,the rest three groups of rats with TNBS/ethanol clyster copy UC rat model.After 7 days,rats in the model group and the blank control group were randomly sacrificed for model evaluation.After the model was successfully replicated,rats in each group were given different drugs with equal volume and dosage by intragastric administration.Rats in the blank control group and model group were given normal saline.Rats in the mesalazine group and sophora powder group were given methalazine solution and Huaihua San decoction respectively.Seven days later,the rats were given abdominal anesthesia,and after anatomy,3-4ml blood was collected from the abdominalaorta of therats for Elisa detection,and about 2cm of diseased colon tissue was removed for histopathological observation and immunohistochemical staining.During the experiment,the rat's spirit,diet,activity,coat,weight and stool were recorded.Record or test results for statistical analysis.Result: Through comparison,it was found that the general condition of UC rats treated with Huaihua San or mesalazine was improved,and the DAI score,colonic histopathological score and CMDI score all decreased,suggesting that the colonic mucosa of UC rats was repaired to different degrees.The levels of in terleukin-6 and transformed growth factor-?1 in serum were higher than those in the blank control group,but decreased significantly compared with the model group.Immunohistochemical staining showed that the expression of intestinal sIgA in rats in the model group was significantly lower than that in other groups,and the expression of intestinal sIgA in rats in the two groups receiving drug intervention was increased compared with that in the model group,but lower than thatin the blank control group.Conclusion: Huaihua San can improve the general disease of UC rats,promote the mucosal repair of UC rats,reduce the level of serum pro-inflammatory factors,reduce the intestinal inflammatory response,improve the expression of intestinal immune protein,and enhance the local immune ability of colon mucosa,which has a good effect on UC rats.The mechanism of action may be through the signaling pathway to regulate the level of inflammatory factors and proteinexpression to play a therapeutic role.
