Objective:To observe the gastric residual rate of the cerebral infarction rats(Middle cerebral artery occlusion,MCAO rats)at different time points(1D,3D,7D,14D after operation)by electroacupuncture at Zusanli and Neiguan.Changes and dynamic changes in the expression levels of connexin 43(CX43),stem cell factor(SCF),and tyrosine protein kinase receptor(c-Kit)in gastric antrum.To explore the potential mechanism of electroacupuncture to improve gastric dysmotility in acute MCAO rats.Methods:120 healthy 8-week-old male SPF SD rats were randomly divided into normal group(group A,n=24),sham operation group(group B,n=24),model preparation 72,model preparation After successful,they were randomly divided into:model group(groupC,n=24),electroacupuncture group(group D,n=24),drug group(group E,n=24);group A,group B,Twenty-four rats in group C,group D and group E were randomly divided into four subgroups:1D,3D,7D and 14D,6/subgroup.Group A did not do any treatment;group B only separated the common carotid artery(CCA),external carotid artery(ECA),internal carotid artery(ICA),not placed in the line.The MCAO rat model was prepared by the Longa modified suture method in the group C,group D and group E.6 hours after the successful preparation of the model,no treatment was given to group A,group B and group C.Electroacupuncture was given to group D(1 time/day),and group E was given mosapride citrate aqueous solution(0.158g)./ml,one at 8 o'clock in the morning and evening).The subgroups were harvested at different time points of 1D,3D,7D and 14D.The abdominal aorta blood and gastric antrum were collected.The general gastric conditions of each group were observed.The gastric residual rate of each group was determined.The levels of motilin MTL and SCF in the serum of abdominal aorta were detected by enzyme-linked immunosorbent assay(ELISA).The gastric antrum tissues of 1D,3D,7D and 14D were observed by HE staining.The pathological changes were detected by RT-PCR.The relative expressions of CX43 mRNA,SCF mRNA and c-Kit mRNA in the gastric antrum of 1D,3D,7D and 14D were detected by immunohistochemistry(IHC).The expression levels of CX43,SCF and c-Ki proteins in gastric antrum tissues of 1D,3D,7D and 14D after operation.Results:(1)Gross observation of gastric tissue of rats in each group:The gastric mucosa folds of rats in group A and group B were rich,the lines were consistent,the color was reddish,the surface was smooth,and no obvious swelling and erosion of gastric mucosa were observed.Compared with group A,rats in group C had more gastric residue at each time point,and the mucosal folds were reduced,the disorder was disordered,the color of the stomach was pale,and the gastric residue of rats in group D,group E and group C was decreased at different time points.The folds are relatively increased,the movement is relatively consistent,and the color of the stomach is slightly reddish.(2)HE pathological observation of gastric antrum tissue of rats in each group:The mucosa of gastric antrum tissue was continuously completed in group A and group B at each time point,the structure was clear,the shape and structure of gland were intact,and a small amount of lymphocyte infiltration was observed in mucosal layer.In group C,the mucosa was almost continuously completed at each time point,and some glandular spaces were slightly widened,glands were dilated,and a few inflammatory cells infiltrated in the mucosa.In group D and group E,the mucosal layer was continuous and complete at each time point,a few glands expanded,and a few inflammatory cells infiltrated in the mucosal layer,but the degree was slightly lower than that in group C.(3)Comparison of gastric residual rate in each group:Compared with group A at the same time,the gastric residual rate of group 1D,3D,7D and 14D was significantly increased in group C,group D and group E(P<0.05).Compared with the group C at the same time,the residual rate of 3D and 7D in the D group and the E group decreased significantly(P<0.05);the residual rate of 3D and 7D in the C group,D group and E group compared with 1D.There was a significant difference in the gradual increase(P<0.05).4)Comparison of serum MTL concentrations in abdominal aorta of rats in each group:Compared with group A at the same time,there was a significant difference in serum MTL concentrations in group 1,D,and 7D between groups C,D,and E(P<0.05).Compared with the C group at the same time,there was a significant difference in the serum MTL concentrations between the D group and the E group in 1D,3D,and 7D(P<0.05).There was a significant difference in the serum MTL concentration in the 3D compared with the 1D.(P<0.05),there was a significant difference in serum MTL concentration between 7D and 14D(P<0.05).There was a significant difference in serum MTL concentration between 7D and 14D in group D and group E compared with 1D(P<0.05).(5)Comparison of serum SCF concentrations in abdominal aorta of rats in each group:Compared with group A at the same time,there was a significant difference in serum SCF concentration in group D at 1D,3D,7D and 14D(P<0.05),group D,There was a significant difference in the concentration of SCF in serum of group E in ID,3D and 7D(P<0.05).Compared with group C in group,there was significant difference in serum SCF concentration between group D and group B in 7D and 14D(P<0.05).Compared with ID,there was a significant difference in serum SCF concentration between 3D,7D and 14D in group C(P<0.05).There was significant difference in 3D serum SCF concentration between group D and group E compared with ID(P<0.05).(6)Comparison of relative expression of CX43 mRNA,SCF mRNA and c-Kit mRNA in gastric antrum of each group:compared with group A at the same time,group C,group D,group E at 3D,7D,14D The relative expression levels of CX43 mRNA,SCF mRNA and c-Kit mRNA in the tissues were significantly different(P<0.05).Compared with the C group at the same time,the CX43 mRNA and SCF mRNA in the gastric antrum tissues of group D and group E at 3D,7D and 14D were compared.There was a significant difference in the relative expression of c-Kit mRNA(P<0.05).Compared with 1D,the relative expression of CX43 mRNA,SCF mRNA and c-Kit mRNA in gastric antrum was decreased in group C.Significant difference(P<0.05);the relative expressions of CX43 mRNA,SCF mRNA and c-Kit mRNA in gastric antrum were significantly different in group D and group E compared with 1D(P<0.05).(7)Comparison of CX43,SCF and c-Kit protein expression levels in gastric antrum tissues of rats in each group:compared with group A at the same time,in group 3,7D,14D,group C,group D,group E CX43,SCF There was a significant difference in the decrease of the IOD value of c-Kit protein expression level(P<0.05).Compared with the C group at the same time,the expression levels of CX43,SCF and c-Kit protein in group D and group E at 3D,7D and 14D were compared.The increase of IOD value was significantly different(P<0.05);the decrease of IOD value of CX43,SCF and c-Kit protein expression levels in 3D,7D and 14D was significant in group C,group D and group E compared with 1D.Difference(P<0.05).Conclusion:(1)There is gastric dysmotility in acute MCAO rats,and the expression levels of CX43,SCF and c-Kit in gastric antrum are decreased.(2)Electroacupuncture at Zusanli and Neiguan can improve gastric motility in acute MCAO rats.The underlying mechanism may be related to up-regulation of CX43 expression,initiation of SCF/c-Kit signaling pathway,and promotion of interstitial cells of Cajal(ICC)Proliferation and improvement of gap junctions.
