Purposes:To explore the intervention effect and mechanism of "Jianpi Huatan,Jiedu hufei" recipe on lung injury caused by PM2.5 from two sources,namely,near surface layer of Nanjing city and coal-fired thermal power plant,so as to provide the therapeutic target and new ideas for PM2.5 prevention and treatment,and provide the scientific basis for clinical application.Methods:Experiment 1:Two PM2.5 particles are collected and analyze for composition by WD-XRF method.Experiment 2:45 rats are randomly divided into blank group,urban model group,power plant model group,urban traditional Chinese medicine high dose group,urban traditional Chinese medicine low dose group,power plant traditional Chinese medicine high dose group,power plant traditional Chinese medicine low dose group,urban western medicine group,and power plant western medicine group.There are 5 rats in each group.Configure the collected two PM2.5 particles into a suspension,and then administer the corresponding groups of rats for 3 weeks;the blank control group was dripped with 0.9%sodium chloride solution through the nasal cavity for 3 weeks from the day the model was established.After the exposure,the city/power plant Chinese medicine low-dose group is given a low-concentration decoction,the city/power plant Chinese medicine high-dose group is given a high-concentration decoction,and the city/power plant western medicine group is given a 7.5%ammonium hydrochloride oral solution.The amount is 3ml/day for each.The blank group and the model group are administrated with saline every day,3ml/day.Dosing is continued for 9 days.After administration,lung tissue sections are prepared.Serum is collected and the expression levels of IFN-y,MDA,AP-1,NF-?B are measured.RNA and total proteins are extracted from lung tissue.HO-1 mRNA,Nrf2,TGF-?,and Smad2 protein levels are detected.Results:Experiment I Results:In urban PM2.5 particle samples,oxygen element(O)accounts for about 44.3%;several metal elements such as calcium,iron,aluminum,magnesium,sodium and potassium account for about 30.64%;non-metal element silicon accounts for about 21.05%and sulfur accounts for about 2.34%.In PM2.5 particles of power plant,oxygen accounts for 96.66%,and a small amount of metal elements and chlorine elements can be detected.The composition of the two PM2.5 samples is quite different.Experiment 2 Results:The levels of AP-1 and MDA in the serum of the two models increase significantly,IFN-? and NF-?B in serum of urban model increase significantly(P<0.05).Comparing with the corresponding model group,the levels of IFN-y,MDA,AP-1,NF-?B in the urban Chinese medicine low-dose group and urban western medicine group,and the urban Chinese medicine high-dose group MDA and AP-1 levels decrease(P<0.05).The serum MDA level in the western medicine group of the power plant is significantly reduced((P<0.01).Comparing with the blank group,the expression levels of Smad,TGF-?,Nrf2,and HO-1 mRNA in the lung tissue of the two model groups are damagingly changed(P<0.01).Comparing with the corresponding model group,each drug group shows different degrees of benign changes(P<0.05).Conclusions:Through Keap1/Nrf2/are,AP-1/Smad/TGF-?,NF-?B and other pathways,Jianpi Huatan-Jiedu Hufei recipe can enhance the antioxidant capacity of the body,inhibit IFN-?-mediated inflammatory damage,and reduce PM2.5 damage to lung tissue.Comprehensive PM2.5 particles in urban areas can damage the respiratory tract through a variety of inflammatory pathways and oxidative stress pathways.The damage mechanism of PM2.5 particles on the respiratory system is mainly caused by oxidative stress mediated by AP-1/Smad/TGF-beta pathway."Jianpi Huatan-Jiedu Hufei" recipe and ambroxol hydrochloride have different intervention effects on the lung injury caused by two PM2.5 particles.Therefore,corresponding treatment plans should be formulated for PM2.5 particles from different sources to achieve curative effects.
