Study On The Material Benchmark Of The Classic Famous Prescriptions Baoyuan Decoction

Objective:Baoyuan Decoction comes from Sun Zhihong's Concise Medicine,which is composed of ginseng,astragalus,licorice,cinnamon,and five Chinese traditional medicines for ginger.It is mainly used to treat the deficiency of vital energy.Based on the prescription dose,medicinal materials,medicinal materials processing and cooking process of the Ming Dynasty in which Baoyuan Decoction is located,the material base preparation method of ancient Classic Prescriptions Baoyuan Decoction was restored,and the quality standard of Baoyuan Decoction material base The preliminary research and the relationship between the magnitude and value transfer will lay the foundation for the development of classic classic preparations in the future.Methods:Based on the research and summary of 369 papers on ancient dose conversion,medicinal herbs and processing,more than 70 ancient Chinese medicine books,and 7 modern monographs,the medicinal herbs in Classic Prescriptions Baoyuan Decoction,their processing specifications and prescription dosages were determined.According to the records of ancient books,combined with modern practical operations,and the Ming Dynasty Baoyuan Decoction material basis preparation method was restored;vacuum freeze-drying was used to dry the decoction.To obtain the material-based lyophilized powder of Baoyuan Decoction.Qualitative identification of the five medicinal materials in the prescription by thin-layer chromatography;using high-performance liquid-phase method with the sum of ginsenoside Rg₁ and Re,ginsenoside Rb₁,and vera isoflavone glucoside as the index components for content determination,and establishing a substance-based quality standards and research on the relationship of quantity value transfer;using fingerprints to identify the characteristic peaks of the five flavor medicinal materials in Baoyuan Decoction and examine the similarity of 15 batches of material benchmarks.Results:Baoyuan Decoction comes from the decoction in"Concise Medicine"from Sun Zhihong of the Ming Dynasty.The medicinal materials and decoction pieces used have been proven to be pharmacopoeial sources and specifications.According to research,the prescription amount in Baoyuan Decoction is:ginseng?3.73g?,astragalus?7.46g?,licorice?1.865g?,cassia?0.746g?,plus a piece of ginger?4-5g?,decoction.The basic preparation method of the reduced substance is:take 4 times the prescription amount,add 1700ml of water?add 100ml water absorption?,soak for 30min,fry for 90-95min,filter with 200 mesh silk cloth,add 400ml of water to the residue,and fry for 25-30min The two filtrates were combined,and the filtrates were left to freeze at-80°C for 24 hours,and then freeze-dried under vacuum for 72 hours.A thin-layer identification method for Wuzhong Fangzhong was established,which was negative and non-interfering.A stable content determination method was established and stipulated that the sum of ginsenoside Rg₁ and Re in the substance standard should not be less than 0.22%,and ginsenoside Rb₁ should not be less than 0.11%,Verbascum isoflavone glucoside must not be less than 0.02%;a material-based fingerprint is established,and the fingerprint similarity of 15 batches of Baoyuan Decoction is>0.90;the value transfer relationship is:Verbascum isoflavone glucoside from Astragalus The average transfer rates of medicinal materials to decoction pieces and decoction pieces to material standards were 110.30%and 59.77%,respectively,and the RSD values were23.40%and 9.44%,respectively;the sum of ginsenoside Rg₁ and Re in ginseng and ginsenoside Rb₁ from medicinal materials to decoctions and pieces The average transfer rates to the substance standard were 103.67%,102.68%,65.03%,68.44%,and the RSD values were 12.31%,15.82%,4.50%,and 8.70%,respectively.Conclusion:Based on ancient classics and literature research,the prescription dosage of Baoyuan Decoction,the basis of medicinal materials,and the processing of medicinal materials were used to restore the preparation method of ancient Baoyuan Decoction.The quality control standards for the material basis of Baoyuan Decoction were initially established.The rule of mass-mass transfer of decoction pieces and material standards provides scientific theoretical basis for the further development and research of the classic Mingfang Baoyuan Decoction.
