Objective: To observe the regulation of Tongxieyaofang on substance P(SP)and neuropeptide Y(NPY)in the colon of 5-HT rats with irritable bowel syndrome(IBS)of liver depression and spleen deficiency syndrome,and to explore the mechanism of Tongxieyaofang in the treatment of IBS of liver depression and spleen deficienc syndrome.Methods: all rats were randomly divided into 4 groups: normal group,model group,traditional Chinese medicine(Tongxieyaofang)group and Western Medicine(pivium bromide)group,with 15 rats in each group.In addition to the normal group,the other groups of rats were made by the method of "restraint + senna leaf gavage".The normal group and model control group were given 0.9% normal saline,western medicine group was given pinaverium bromide,Tongxieyaofang group was given Tongxieyaofang solution for treatment.After the treatment,the content of 5-HT,SP and NPY in hypothalamus and colon were measured by immunohistochemistry.Tukey multiple comparison test of graphpad prism version 7.0 statistical software was used to analyze the experimental data,and one way ANOVA was used to analyze the differences between groups.Results: Compared with the normal group,the expression of 5-HT in the hypothalamus of the model group decreased(p<0.01);Compared with the model group,the expression of 5-HT in the hypothalamus of Tongxieyaofang group increased(p <0.001);the expression of 5-HT in the hypothalamus of Pinaverium group increased,but the difference was not statistically significant.Compared with the normal group,the SP immunohistochemical staining intensity of the colon in the model group was significantly higher(p <0.0001);compared with the model group,the SP immunohistochemical staining intensity of the colon in the Tongxieyaofang group was significantly lower(p<0.0001),and the SP immunohistochemical staining intensity of the colon in the Pinaverium group was significantly lower(p< 0.01);There was no significant difference between the two groups.Compared with the normal group,the NPY immunohistochemical staining intensity of colon in the model group was significantly lower(p< 0.0001);Compared with the model group,the NPY of colon in Tongxieyaofang group and pivealammonium group increased(p< 0.0001);There was no significant difference between the two groups.Conclusion:(1)The central control center of Tongxieyaofang in the treatment of IBSwith liver depression and spleen deficiency may be located in the hypothalamus.The mechanism of relieving the symptoms of liver depression is related to the increase of 5-HT content in the hypothalamus.(2)The mechanism of Tongxieyaofang in soothing the liver and relieving pain may be as follows: It can reduce the secretion of SP in colon and inhibit the transmission of pain information by SP.It can increase the content of NPY in colon,inhibit the excitation of primary afferent neurons,and reduce the production of pain signals.(3)The mechanism of Tongxieyaofang in strengthening the spleen and relieving diarrhea may be: Reducing the secretion of SP in colon,improving gastrointestinal motility and reducing mucus secretion.Increasing the secretion of NPY in colon,inhibiting the secretion of intestinal fluid and pancreatic fluid,and inhibiting gastrointestinal motility.
