Experimental Study Of Gastrodiaand Uncaria Decoction With Acupuncture At Taichong Point On Spontaneously Hypertensive Rats

Objective: Through the experimental study of Gastrodia and Uncaria Decoction and Taichong Point Acupuncture on spontaneous hypertension rats,to verify whether the antihypertensive effect is synergistic,and to explore the possible mechanism of antihypertensive effect on spontaneous hypertension rats.Methods: Forty spontaneously hypertensive rats were randomly divided into model group,Taichong group,traditional Chinese medicine group and combined group,10 rats in each group.The corresponding treatment was carried out on Taichong group,traditional Chinese medicine group and combined group every day.After 4 weeks,the serum and renal tissue of abdominal aortic blood were taken,and the NPY of renal tissue was amplified by RT-q PCR The expression of NPY gene was calculated by2-△△ct method,the concentrations of S-cr,BUN,NPY,ET-1,CGRP and NT in serum of rats were detected by ELISA,the morphological changes of renal tissue were observed by HE staining,and the data were statistically analyzed by SPSS 24.Results: Compared with the model group,the blood pressure of the combined group,the traditional Chinese medicine group and the Taichong group decreased significantly(P <0.05);compared with the traditional Chinese medicine group and the Taichong group,the blood pressure of the combined group was statistically different(P < 0.05);S-cr and BUN were significantly decreased in the combined group,the traditional Chinese medicine group and the Taichong group(P < 0.05),and there was no significant difference between the Taichong group and the traditional Chinese medicine group(P >0.05)NPY in the serum and renal tissue of the combined group,the traditional Chinese medicine group and the Taichong group were lower than that of the model group,and there was a statistical difference(P < 0.05),among which the concentration of NPY in the combined group was higher than that of the traditional Chinese medicine group and the Taichong group(P < 0.05);the level of CGRP and NT in the serum of the Taichong group,the traditional Chinese medicine group and the combined group were higher than that of the model group There was no significant difference in CGRP expression among the three groups(P > 0.05),NT level between the two groups(P < 0.05),and there was no significant difference between the two groups(P > 0.05);serum ET-1 level of the three groups was significantly lower than that of the model group There was no difference between Taichong group and traditional Chinese medicine group(P > 0.05).Compared with the control group,the renal tissues of the combined group,the traditional Chinese medicine group and the Taichong group were more complete,and the atrophy and hyperemia of the blood vessels were improved.Conclusion: Gastrodia and Uncaria Decoction combined with acupuncture has therapeutic effect on hypertension in spontaneously hypertensive rats,and the effect is better than that of medication alone or acupuncture.It has a certain improvement on renal damage caused by hypertension.Its mechanism may be related to the reduction of NPY and ET-1 levels in vivo,and the improvement of CGRP and NT level.
