Coix is a medicinal and edible plants,it contains a variety of nutrients.The research at home and abroad has shown that it contains lipids,triterpenoids,alkaloids,polysaccharides and other compounds.Its extracts have antipyretic,analgesic,sedative,anti-tumor,anti-inflammatory,hypoglycemic and lipid-lowering and other effects.Among them,Coixan has the functions of hypoglycemic,lipid-lowering and anti-tumor and other effects.However,there is a lack of research on the molecular mechanism of its action.In this study,Coixan and Coix seed powder were used to study the hypoglycemic and lipid-lowering effects of mice.Through the comparative analysis of various physiological indexes of mice and sequencing of liver transcriptome,the aim of this study is to reveal the mechanism of action of Coixan on glycolipid metabolism.High-carbonhydrate experiment:Diabetic mice were modeled by high sugar feed and streptozotocin.The experimental results show that diabetic mice have the characteristics of polydipsia,polyphagia,and weight loss.Coixan increases the weight of the pancreas in diabetic mice and normal mice.It can be seen from pathological sections that Coixan has a repairing effect on liver damage.Among the indicators measured by serum,Coixan can reduce the area under the curve(AUC)of oral glucose tolerance test in diabetic mice.It can significantly reduce the content of T-CHO,LDL-C,HDL-C,GPT,MDA,SOD,increase the content of TG,GOT.By measuring liver tissue,it can show that Coixan can reduce the content of T-CHO.TG GOT and MDA in normal mice,and increase the content of GPT.It can reduce the content of T-CHO,GPT.GOT.MDA and SOD in diabetic mice and increase the content of TG.The transcriptome sequencing analysis of liver tissue showed that Coixan had significant effects on the main genes of glycolipid metabolism,such as Lpl,Grb10,Gstal and Abcgl,and further verified the quantitative PCR of its gene,It was confirmed that Coixan has a more significant down-regulation of gene expression levels in mice after blood glucose elevation.High-fatty experiment:Modeling of hyperlipidemia mice is performed by high-fat diet.Coixan and Coix seed powder had no significant effect on body weight,diet and drinking water of mice.However,Coixan could increase the weight of liver.Liver pathological sections showed that Coixan had a repairing effect on liver damage caused by hyperlipidemia.Among the indicators measured by serum,Coixan can decrease the content of T-CHO,GOT,MDA and SOD,and increase the content of TG,but Coix seed powder can increase the content of HDL-C,and has no significant effect on other indicators.Coixan and Coix seed powder can reduce the content of serum glucose.Through the physiological indicators measured in the liver.Coixan can reduce the content of T-CHO.TG,GPT,increase the content of LDL-C,SOD,GOT,MDA.Coix seed powder can reduce the content of T-CHO,TG,increase the content of LDL-C,GOT,MDA,Sequencing of liver transcriptome sequencing and validation of related gene-quantitative PCR showed that the major genes involved in glycolipid metabolism after Coixan treatment:The expression levels of related genes such as Acot3 and Sikl were significantly up-regulated,and the expression levels of related genes such as Mup17 and Gstal were significantly down-regulated.This indicates that Coixn has a certain effect on reducing blood glucose in diabetic mice and hyperlipidemia mice.It has certain lipid-lowering and anti-oxidant effects on normal mice,diabetes mice and hyperlipidemia mice.And it has a certain ability to repair liver damage.Coix seed powder has only a certain hypoglycemic and lipid-lowering effect on hyperlipidemia mice.They achieve the effects of hypoglycemic and lipid-lowering by regulating the metabolic pathways involved in glycolipid metabolism.
