Type2diabetes is a genetic metabolic diseases, a serious threat to human health has become one of the major chronic diseases of modern society. Rodent as an important Animal model of type2diabetes, pig models are also reported, but the research is relatively small. The structure of physiological, anatomical, physiological and biochemical index, food structure and drug metabolism between pigs and human are high similarity. Pigs are one of the ideal animal model for human medical research. Type2diabetes a genetic and phenotype are highly heterogeneous complex. Disease-related genes and the interaction have significance in elucidating the pathogenesis and pathological of the type2diabetes, prevention, and finding effective treatment methods.In this study, high-fat high-sugar diet induced Bama miniature pigs22month, indicators of body weight, blood glucose, impaired glucose tolerance, insulin, and insulin resistance index about the type2diabetes each month had changed; throughout the induction process. These biochemical indicators of changes have important role in understanding the disease process. Liver RNA sequencing, transcriptome analysis of live related gene expression and the interaction between process-related indicators to type2diabetes, and the interaction between genes indicating the mechanism of type2diabetes.Relevant indicators of Bama miniature pigs induced by high-fat high-sugar diet long-term process:body weight, insulin, insulin resistance index, glucose tolerance, blood indicators of total protein, serum total cholesterol, LDL, HDL, blood urea nitrogen, creatinine, etc. significant changed, indicating reaction of type2diabetes process-related physiological.High fat and sugar long-term induction of miniature pigs, a variety of blood biochemical parameters changed,13kinds of cytokines including SOD and serum levels of resistin changed.High-throughput liver RNA sequencing of Miniature pig are using for analysis the expression of related genes and the interaction. The results of sequencing Were identified by quantitative real-time quantitative PCR.After induction the main differences of the miniature pig liver genes GO analysis found that the experimental group and control group differences genes distribution in the three GO Term are the biological process GO, cellular component GO and molecular function GO.Liver main differences genes distribution are the fatty acid metabolic pathways and cellular inflammatory pathways. The fatty acid metabolic pathways in miniature pig liver include fatty acid fatty acid desaturase, extend the carbon chain and triglyceride synthesis. Inflammation-related genes analysis showed that the inflammation-related genes of the experimental group were fully raised, these immune-related pathways raised in the miniature pig liver showed that the experimental group of oxidative stress and inflammation inflammatory response.Through the study we got insulin resistance, hyperinsulinemia, impaired glucose tolerance, miniature pigs suffering from fatty liver and hepatitis disease, type2diabetes; analysis of the relevant indicators of type2diabetes, weight, glucose tolerance, blood glucose and insulin analysis of gene expression and the relationship between the level of liver transcriptome. This will be of great significance for the mechanism of type2diabetes, prevention and treatment of disease. |
