Objective: To explore the relationship between the polymorphism of NFKB1 and I?B? gene and the risk of lung cancer,and to explore the interaction between NFKB1 and I?B? gene polymorphism and environmental factors on the risk of lung cancer.Methods: A casecontrol study was carried out.The case group is the primary lung cancer group admitted to the Third Affiliated Hospital of Xinjiang Medical University,and the control group was a group of 150 healthy patients who came to the hospital for physical examination during the same period.General information about the population,such as age,lifestyle and family history,was collected using the Chinese urban cancer screening questionnaire.1ml peripheral venous blood was harvested from each sample and analyzed by RFLP-PCR.Genotyping of rs28362491,rs3774963,rs696 and rs3138053 was performed.Results: After adjusting for age,education level and occupation,multivariate regression analysis showed that Consumption of fruit greater than 1.25kg/week(OR=0.093,95%CI:0.010-0.841,P=0.034),drinking tea(OR=0.099,95%CI:0.042-0.232,P=0.000)and reducing the risk of lung cancer Relevant;edible animal meat is greater than 350 g /week(OR=4.208,95%CI:1.761-10.056,P=0.001),kitchen fume is more(OR = 4.863,95%CI: 2.225-10.631,P=0.000),smoking is greater than 20 years(OR=7.476,95% CI: 1.543-36.223,P=0.011),high blood lipid(OR=6.943,95%CI: 4.861-32.801,P=0.000),high blood lipid(OR=6.943,95%CI:1.264-38.123,P=0.043)may promote the occurrence of lung cancer.NFKB1-rs28362491,rs3774963 and I?B? disease-rs3138053 polymorphisms showed statistically significant differences in lung cancer risk between the case group and the control group.The results of gene-gene interaction showed that the model(rs28362491,rs3774963,rs696,rs3138053)was the optimal model.In the gene-environment interaction,the model(rs28362491,rs3774963,rs696,rs3138053,fruit consumption,smoking,tea drinking,mental stress,pulmonary disease history,hyperlipidemia)was the optimal model.Conclusion: NFKB1 rs 28362491,rs3138053 and I?B? positive rs3138053 were associated with lung cancer risk in NF-?B pathway,and no association was observed between I?B? positive rs696 and lung cancer.
