| Objective:To study the changes of intestinal microflora structure,serum inflammatory factors IL-6,IL-23 and IL-10,and the relationship between the microflora structure and inflammatory factors in patients with colorectal cancer,so as to infer the possible pathogenesis of colorectal cancer and provide new guidance for its treatment.Methods: 1.According to the pre-set inclusion and exclusion criteria,39 patients with colorectal cancer in the First Affiliated Hospital of Xinjiang Medical University were randomly sampled,including 1g of morning stool and 5ml of peripheral venous blood.DNA and serum inflammatory factors were extracted from patients’ faeces as the experimental group samples,and 30 healthy people’s faeces DNA and serum inflammatory factors as the control group samples.2.According to the instructions of the relevant kit,obtain the DNA of the two groups of people’s morning stool flora(total intestinal flora,Bacteroides,Clostridium perfringens,Escherichia coli,Citrobacter,Lactobacillus,Bifidobacterium),and detect the number of copies of the flora by real-time fluorescent quantitative PCR.3.The serum levels of IL-6,IL-23 and IL-10 were measured by enzyme-linked immunosorbent assay(ELISA).4.Using SPSS 20.0statistical software to test and analyze the correlation.Results: 1.Compared with the healthy control group,the total number of copies of intestinal flora in the experimental group decreased,and the average number of copies of Bacteroides,E.coli,and Clostridium perfringens increased significantly(P < 0.01);the number of probiotics bifidobacteria and Lactobacillus in the experimental group decreased significantly(P <0.01);the average number of copies of Citrobacter increased High,but p > 0.05,no statistical significance.2.The average content of IL-6 in the experimental group was significantly higher than that in the control group,P < 0.01,with statistical significance;the average content of IL-23 was higher than that in the control group,P < 0.05,withstatistical significance;the content of IL-10 was decreased,P < 0.01,with statistical significance.3.The average copy number of Bacteroides,Escherichia coli and aerocapsular bacteria showed a positive correlation(r > 0)with IL-6 and IL-23,and a negative correlation(r < 0)with IL-10;there was no significant correlation between Citrobacter and IL-6,IL-23,IL-10;there was a negative correlation(r < 0)between Bifidobacterium,Lactobacillus and IL-6,IL-23,and a positive correlation(r > 0)with IL-10.Conclusion:1.The change of intestinal microflora structure in CRC patients is reflected in the change of species and quantity of microflora,the decrease of beneficial bacterial copies and the increase of harmful bacteria.2.2.The levels of IL-6,IL-10 and IL-23 in the peripheral venous blood of the two groups were detected by ELISA.The level of IL-6 and IL-23,which can promote the inflammation,increased in colorectal cancer patients,and the level of IL-10,which can inhibit the inflammation,decreased in CRC patients.3.The change of intestinal flora structure of CRC is related to the occurrence of inflammatory immunity,which provides a new channel for clinical treatment. |
