Objective: Application of percutaneous coronary intervention(PCI),to investigate mixed thrombus what in vitro simulated making placed into the canine coronary artery,to establish an experimental model of canine acute coronary occlusion.Methods: Fourteen healthy Beagle dogs aged 10-13 months were randomly selected.The dogs were randomly divided into two experimental groups according to the length of occlusion time.The postoperative 2h group(group 1)and the postoperative 4h group(group 2)),7 in each group.Intravenous venous blood(VB)was taken 10 ml/s before operation,and mixed thrombus was prepared in vitro.The mixed thrombus was placed into the coronary artery through PCI to establish an animal model of acute coronary occlusion in dogs.Postoperative group 1 and group 2 underwent routine Electrocardiogram(ECG)and Troponin T(cTnT)examination;Echocardiographic LVESV,LVEDV,LVEF and FS datas;Myocardial Contrast Echocardiography(MCE): The values of myocardial perfusion parameters(A,?,and MBF(A × ?))were determined using Q-lab(Philips Ultrasound,version 10.8);after sacrifice,the myocardium was stained with HE.Results:A mixed thrombus mainly composed of fibrin mesh and activated platelets was prepared successfully in vitro by dog's VB.After the mixed thrombus was placed into the coronary artery,the large digital subtraction angiography(DSA)showed no perfusion(TIMI 0),it confirmed the coronary completed occlusion.ECG showed all dogs the ST segment elevation;cTnT did not increase in group 1 compared with preoperative,but it increased in group 2(P <0.05).Echocardiography and MCE showed that LVEF,FS and MBF values in group 1 and group 2 were lower than before surgery(P<0.05);LVEDV andLVESV values were increased after surgery compared with preoperative.In the HE staining of group 1 and group 2 after surgery,the myocardial cells were arranged neatly,no destruction and necrosis of cardiomyocytes.Conclusion: Our findings demonstrate that the dog's VB can be successfully simulated in vitro to prepare the mixed thrombus.Using of PCI to place a mixed thrombus into the canine coronary artery can successfully establish an experimental model of acute coronary occlusion.
