Effect Of Aconitum Diphtheriae On Th1?Th2 Related Cytokines In Collagen-induced Arthritis Rats

Objective:To study the effects of diphtheria aconitum on Th1 ? Th2 related cytokines in collagen-induced arthritis(CIA)rats,further study and discuss the mechanism of diphtheria aconitum on rheumatoid arthritis.Methods: Healthy 6-8 week-old DBA/1 male rats,weighing 18-22 g,4mg/ml bovine type II collagen(CII)and complete Freund's adjuvant(CFA)are fully dosed on ice at 1:1 emulsification.The primary immunization will be performed,and 21 days after the initial immunization,the foot cream and tail roots will be boosted again with the emulsion described above.On the 35 th day after the boost,the swelling progress was maximized and the changes in mental activity of the rats were also inspected at the same time.Each group of rats is scored every week.After the modeling is completed,they are randomly divided into normal group,model group(collagen-induced arthritis group),tripterygium group,diphtheria aconitum group,normal control group and collagen-induced arthritis.Groups were given gavage at a 1:1 dose of normal saline.The body weight of the rats was measured every week,and the dose was adjusted according to the weight change.After modeling,each rat in the tripterygium and diphtheria aconitum groups was given the corresponding test drug daily.The normal control group and the CIA model group were given the same amount of physiological saline,and continued to be administered for 4weeks.ELISA Test(ELISA)to detect the levels of IL-2?IL-5?IL-13?IFN-??TNF-?.Results: Compared with the model group,diphtheria aconitum and tripterygium polyglycoside can significantly improve the severity of RA in CIA rats compared with the model group.From the experimental point of view,the serum IL-2 ? IL-5 ? IL-13 ? IFN-? ? TNF-? concentration levels were significantly reduced(P<0.05).Conclusion: Combined with the aboveexperimental results,the diphtheria aconitum regulates Th1 ? Th2 cells in collagen-induced arthritis,resulting in immune dysfunction,it plays an important role in rheumatoid arthritis in clinical treatment.
