Objective:To investigate the effect of exercise intervention on inflammatory cytokines,adipokines and endothelial dysfunction(ED)in diet-induced obese mice,and explore the possible mechanism of exercise improving endothelial dysfunction in obese mice,so as to lay the foundation for the targeted study of the "dose-effect relationship" between exercise intervention and prevention and treatment of endothelial dysfunctionMethod:41 SPF C57BL/6 mice(20 males,21 females)were randomly divided into normal control group(NC,n=11,5 males,6 females)and high-fat diet group(HFD,n=30,15 males,15 females),separately fed ordinary food and high fat diet.After 9 weeks,25 obese mice were randomly divided into obesity exercise group(OE,n=13,5 males,8 females)and obesitycontrol group(OC,n=12,5 male,7 females),continued to feed on high fat diet.The OE group was trained for 8 weeks,without weight-bearing swimming,60min per time,1 time per day,6 days per week.After 8 weeks swimming intervention,measure their weight and the APN,Omentin,Leptin,Visfatin,CRP,IL-6,TNF-?,sTM in serum were measured by ELISAResult:1.There was no significant difference in the gengeral data of weight between NC and HFD before making models(P>0.05).There was no significant difference in the gengeral data of weight between mice in OE and OC before exercise intervention(P>0.05)2.Compared with NC,the weight of mice in both OC and were increased(P<0.01).Compared with OC,mice in OE's weight were decreased(P<0.05)after 8-week swimming exercise intervention3.? Compared with NC,the sreum sTM levels in HFD increased significantly before exercise intervention(P<0.01).?After 8-week swimming exercise intervention,compared with NC,the serum Leptin,Visfatin,CRP,IL-6,TNF-?and sTM levels in OC increased significantly(P<0.01);the serum APN and Omentin decreased,but the difference of them were not significant(P>0.05).Compared with NC,the serum Omentin levels in OE increased significantly(P<0.05);the serum APN increased,but the difference of them were not significant(P>0.05);the serum Visfatin,CRP,IL-6,TNF-?and sTM levels in OE increased significantly(P<0.01);the same as the serum Leotin(P<0.05).Compared with OC,the serum APN levels(P<0.05)and the serum Omentin levels(P<0.01)of the OE increased;the levels of Visfatin of OE decreased(P<0.05);the levels of Leptin,CRP,IL-6,TNF-?and sTM of OE decreased significantly(P<0.01)4.There was a intermediate negatively correlation between serum APN levels and weights in mice(P<0.05).Serum Omentin levels were strong negatively correlated with weights(P<0.01).Serum CRP levels were intermediate positively correlated with weight(P<0.05).There was a weak negative correlation between levels of serum APN and serum IL-6 in mice(P<0.05)Serum APN levels were intermediate positively correlated with serum Omentin levels in mice(P<0.05).Serum Omentin levels were intermediate negatively correlated with serum CRP levels(P<0.01).Serum Leptin levels were intermediate positively correlated with both levels of serum IL-6 and serum TNF-?(P<0.01),the same as serum Visfatin levels(P<0.01).Serum Visfatin levels were strong positively correlated with serum TNF-alevels in mice(P<0.01)Additionally,there was a extremely strong positively correlation between serum IL-6 levels in mice(P<0.01).Serum CRP levels were intermediate positively correlated with serum sTM(P<0.01).Both serum Leotin levels and serum IL-6 levels were strong positively correlated with serum sTM levels(P<0.01).Both serum Visfatin levels and serum TNF-alevels were extremely strong positively correlated with serum sTM levels(P<0.01)Conclusion:1.Obesity leads to inflammatory response and imbalance of adipokines,which affects the occurrence and development of ED2.8-week swimming exercise intervention can be used as a method of weight loss3.The 8-week swimming exercise intervention can effectively reduce the inflammatory response in obese mice,which may be related to its stimulation of the secretion of APN,Omentin,and the inhibition of the secretion of pro-inflammatory factors Leptin and Visfatin.Thus,endothelial dysfunction can be relieved by inhibiting the inflammatory response and maintaining the balance of adipokines.
