ObjectiveTo explore the ablation mechanism and therapeutic effect of a new type of high-frequency wave pulse in the treatment of liver cancer in rats,and to provide a basis for exploring a new method of tumor ablationMethodsMcA-RH7777 cells were cultured in vitro to prepare tumor suspension.Clean grade SD rats were injected intrahepatic to establish liver cancer model.One week after modeling,40 rats were randomly divided into experimental group(n=26)and control group(n=14).In the experimental group,the liver tumor was treated with electric pulse under the field strength of 2000 V/cm,while in the control group,the pulse field strength was 0 V/cm.The changes of serum biochemical indexes,Myocardial Zymogram and AFP level of tumor markers in the two groups were monitored 3 days,1 day before and 1 day,7 days,14 days,21 days and 30 days after the treatment.The changes of tumor length and diameter,tumor volume and survival time of 60 days before,7 days,14 days and 21 days after the operation were recorded respectively.The complications during the treatment were recorded at random 30 days after the operation The expression of Caspase-3,Bcl-2 and Bax protein in the ablation target area of the two groups were detected,and the ablation area of the tumor was cut for HE staining,TUNEL staining,Ki67 and VEGF immunohistochemistry to observe the histomorphology change,apoptosis rate and cell proliferation rateResultIn the experimental group,the serum ALT and AST were increased one day after operation,and then decreased continuously.The serum ALT was close to the normal level 21 days after operation,and the serum ALT and AST in the control group were increased continuously.There was no significant difference in creatinine and urea nitrogen between the experimental group and the control group before and after the experiment.Serum phosphokinase and LDH were increased one day after operation,and then decreased continuously.There was no significant difference between the two groups at each time point.AFP in the experimental group reached the peak on the first day after operation,showing a continuous downward trend,and reached the normal value on the fourteenth day after operation AFP in the control group continued to rise,and began to show a difference on the seventh day after operation,and the difference was statistically significant.Western blot showed that the expression of Caspase-3 and Bax increased and Bcl-2 decreased in the observation group.In the experimental group,the tumor length and diameter were(11.2±3.1,8.1 ± 2.3,5.3 ± 1.6,3.5± 1.1)mm on the first day,the seventh day,the fourteenth day and the 21st day,respectively.The tumor volume was(301.8±243.0,139.7 ± 128.1,45.0 ± 39.5,13.5±12.5)mm3.In the control group,the tumor length and diameter were(9.8±2.1,14.± 2.7,17.8 ±3.7,14.4 ± 2.7)mm,respectively.The volume was(267.6 ± 192.6,736.3±391.9,1488.8 ± 671.7,901.3 ± 428.0)mm3,which showed a continuous downward trend,reached the maximum value 14 days after operation,and slightly decreased 21 days after operation.In the experimental group,he staining showed a large number of apoptotic and necrotic areas,with clear boundary with normal tissue.There were fragmented cells and inflammatory cells in the necrotic area,and the ductal system remained intact without obvious damage.VEGF immunohistochemistry showed that the number of blue staining cells in the experimental group was significantly less than that in the control group,TUNEL staining showed strong staining in the experimental group,but weak staining or no staining in the control group,theapoptosis rate in the experimental group was 76.85 ± 10.27%,and the apoptosis rate in the control group was 2.56±1.67%.The proliferation rate of Ki67 was 2.8±0.8%.In the control group,the proliferation rate was 43.1 ± 3.8%.Conclusion the new type of UHF square wave pulse can effectively control the local progression of liver cancer in rats,and it has a good effect.Conclusion1.The McA-RH7777 cell liver tumor model prepared by direct injection has a high tumor formation rate and low cost,and is a very simple and reliable method for preparing liver cancer models.2.The high-frequency square wave electric pulse can effectively control the local progression of rat liver cancer,effectively protect normal vascular tissue,and has good safety and effectiveness.3.The new high-frequency square wave pulse can effectively reduce complications such as muscle contraction and arrhythmia.
