ObjectiveTo detect the expression of FOXC2 and vasculogenic mimicry in epithelial ovarian cancer tissue,analyze the correlation between them.And to investigate the effect of RNA interference-based silencing of FOXC2 gene on the expression of VE-cadherin and vasculogenic mimicry,to explore the effect of FOXC2 in epithelial ovarian cancer development and metastasis.Methods1.Detect the expression of FOXC2 in normal ovarian tissue and epithelial ovarian cancer tissue by immunohistochemical method.and to explore if VM exist in epithelial ovarian cancer tissue by double staining with CD34 and PAS,to investigate the correlation between the expression of FOXC2 and VM in epithelial ovarian cancer.2.FOXC2 siRNA and N-control siRNA were transfected into ovarian cancer cell line SKOV3.3.RT-PCR method were applied to detect the expressions of FOXC2 mRNA in three groups.4.A well-established model of Three-dimensional Matrigel culture in vitro was used to compare the ability to form vascular mimicry(Vasculogenic mimicry,VM).Results1.The expression of FOXC2 in normal ovarian tissue was negative or weakly positive,but in epithelial ovarian cancer tissue the expression of FOXC2 was strongly positive.The expression rate of FOXC2 in epithelial ovarian cancer tissue was higher than that in normal ovarian tissue,the difference was statistically significant(P<0.05).2.The expression level of FOXC2 was related to the epithelial ovarian cancerstage(P<0.05),and have no relation with the patient's age,histological type,differentiation status and lymph node transfer.3.The expression rate of VM in epithelial ovarian cancer tissue was higher than that in normal ovarian tissue,the difference was statistically significant(P<0.05).4.The expression rate of VM was related to the differentiation status and lymph node transfer(P<0.05),and have no relation with the patient's age,histological type and cancer stage.5.The expression rate of VM was positively correlated with the expression of FOXC2(r=0.404).6.RT-PCR verified that FOXC2-interference SKOV3 cell line was established successfully.RT-PCR results showed that the expression of FOXC2 mRNA level was decreased in FOXC2-interference group,at the same time,the expression of VE-cadh erin mRNA also showed a trend of decrease,the difference was statistically significant(P<0.05).7.Matrigel angiogenesis method showed that FOXC2 interference decreased the number of tube formation of SKOV3 cells,the difference was statistically significant(P<0.05).Conclusion1.FOXC2 may be effected the development of epithelial ovarian cancer.2.VM was exisited in the epithelial ovarian cancer,the high expression of FOXC2 maybe promote the formation of VM.3.The high expression level of FOXC2 and VM may be related to the progressi on and metastasis of epithelial ovarian cancer.4.Silenced the expression of FOXC2 by siRNA interference technology,the expression of VE-cadherin was decreased,and the ability of tube formation was reduced,suggested that targeting silence the expression of FOXC2 maybe could reduce the VM.
