Objective:prostate cancer is one of the most common malignant tumors in men.This paper discusses the correlation between Pokemon expression in prostate cancer and clinical parameters,and provides a theoretical basis for future treatment of prostate cancer.Methods:real-time fluorescence quantitative PCR(RT-PCR)was used to detect the mRNA expression of Pokemon gene in prostate cancer and benign prostatic hyperplasia tissues,including 30 cases of prostate cancer as the experimental group and 30 cases of benign prostatic hyperplasia as the control group.We analyzed the expression of Pokemon in cancer group compared with the control group and its correlation with clinical stage,preoperative PSA level,pathological Gleason score,age,lymph node metastasis,seminal vesicle invasion,surgical margin and Magnetic Resonance Imaging(MRI)results.Immunohistochemical method was also used to detect the expression of Pokemon in prostate cancer and benign prostatic hyperplasia specimens.The experimental group was 47 paraffin tissue specimens of prostate cancer,and the control group was 30 cases of prostatic hyperplasia paraffin tissue.Furthermore,we correlated its expression level with the cumulative survival rate and tumor biochemical recurrence-free survival rate using the follow-up data among the prostate cancer patients.Results:The expression of Pokemon mRNA in prostate cancer was higher than that in benign prostatic hyperplasia,and there is significant difference(P <0.05).The expression level of Pokemon in prostate cancer was correlated with clinical stage,preoperative PSA level and pathological Gleason score(P<0.05).There was no significant correlation with age,lymph node metastasis,seminal vesicle invasion,surgical margin and MRI(P>0.05).Immunohistochemistry results showed that Pokemon was mainly expressed in the nucleus of prostate cancer cells.The staining showed brownish yellow.Among 47 cases of cancerous tissues,22 were highly expressed,accounting for 46.8%(22/47),while 30 cases of benign prostatic hyperplasia showed low expression.The comparison between the two groups was statistically significant(P<0.05).The expression of Pokemon in prostate cancer was significantly higher than that in benign prostatic hyperplasia,which was consistent with the conclusion of RT-PCR.By observing the expression of Pokemon via immunohistochemical method in postoperative specimens of prostate cancer,there was no significant correlation between the expression of Pokemon in prostate cancer and the postoperative cumulative survival rate(P>0.05).But there was significant correlation between the expression of Pokemon in prostate cancer and biochemical recurrence-free survival rate(P<0.05).Conclusion:1.The mRNA and immunohistochemical expression of Pokemon in prostate cancer tissue were higher than that in benign prostatic hyperplasia tissue.2.The expression of Pokemon in prostate cancer tissue was significantly correlated with clinical stage,preoperative PSA level and Gleason score.There was no correlation between Pokemon expression and Age,lymph node metastasis,seminal vesicle invasion,surgical margin,MRI.3.The expression of Pokemon in prostate cancer was statistical significance in the terms of biochemical recurrence-free survival rate.There was no significant correlation between the expression of Pokemon and the cumulative survival rate in prostate cancer.The biochemical recurrence free survival rate of the over expression patients was lower than that of the low expression patients.
