Objective:To explore the expression of HIST1H3 D gene in normal cervical tissues,cervical intraepithelial lesion tissues and cervical cancer tissues,and further analyze the relationship between the expression of HIST1H3 D gene in cervical cancer tissues and the clinicopathological characteristics and pelvic metastasis.Methods: Collected the pathology paraffin block specimens and surgical treatment in the Department of Gynecology in the First Affiliated Hospital of Bengbu Medical College,during February 2012 and December 2013,including 20 cases of low-grade cervical intraepithelial lesions,20 cases of high-grade cervical intraepithelial lesions,and 74 cases of cervical cancer.During the same period,hysterectomy was performed for uterine fibroids and 20 paraffin specimens were pathologically diagnosed as normal cervical tissue.Immunohistochemistry was used to detect the expression and differences of HIST1H3 D gene in normal cervical,low cervical intraepithelial lesion(LSIL),high cervical intraepithelial lesion(HSIL)and cervical cancer tissues,to collect information and clinical data of patients,and analyze the relationship between the expression of HIST1H3 D gene and the clinicopathological features and pelvic metastasis of cervical cancer.Results : 1.The expression of HIST1H3 D gene in cervical cancer tissues was significantly higher than that in normal cervix,low cervical intraepithelial lesion and high cervical intraepithelial lesion,with statistically significant difference among the four groups(P <0.01).Moreover,compared with the four groups,the expression of HIST1H3 D gene in cervical cancer tissues was statistically significant in normal cervical tissues,low cervical intraepithelial lesions(LSIL)and high cervical intraepithelial lesions(HSIL)(P <0.05).There was no statistically significantdifference between HIST1H3 D gene expression in normal cervical tissue and LSIL(P >0.05).There was no statistically significant difference between HIST1H3 D gene expression in LSIL and HSIL(P >0.05).2.Relationship between HIST1H3 D gene expression in cervical carcinoma and clinicopathological features: The differences of HIST1H3 D expression in FIGO staging,pathological grading and lymph node metastasis were statistically significant(P <0.05).The expression in lymph node positive cervical cancer tissues was higher than that in lymph node negative cervical cancer tissues.The higher the FIGO staging was,the higher the HIST1H3 D expression was,and the difference was statistically significant(P <0.05).The lower the pathological differentiation was,the higher the HIST1H3 D expression was,and the difference was statistically significant(P<0.05).However,it was not correlated with age,pathological classification and tumor size(P >0.05).Conclusion: 1.The expression of HIST1H3 D gene in normal cervical tissues,low grade intraepithelial lesion,high grade intraepithelial lesion and cervical cancer tissues increased gradually,which may be related to the occurrence and development of cervical cancer.2.The expression of HIST1H3 D gene in cervical cancer was negatively correlated with the degree of pathological differentiation,and positively correlated with clinical stage and lymph node metastasis,which may serve as a reference indicator for lymph node metastasis and malignancy of cervical cancer.
