Objective.By constructing a short hairpin RNA(shRNA)that knocked down the HS3ST2 gene from mice,and transfected into the rat Growth Hormone-Secreting Pituitary Adenoma GH3 cell line,further study the role of the gene HS3ST2 in GH adenoma through cell experiments The influence of genes on the growth and proliferation,apoptosis and hormone secretion of GH adenoma.Methods.We constructed the short hairpin RNA(shRNA)of the murine knockdown gene HS3ST2,which was transfected into the GH3 cell line in GH adenomas of rats.The expression levels of gene HS3ST2 in GH3 cells before and after transfection were tested through RT-qPCR.The cck-8 method was used to measure the viability of GH3 cells in each group at 24 h,48h,72 h,and 96 h after transfection.72 hours after transfection,the secretion levels of GH and IGF-1 in the supernatants of GH3 cells in each group were detected by ELISA.The relative levels of apoptosis of GH3 cells in each group after 72 h of transfection were detected by flow cytometry.Results.When the expression of HS3ST2 in GH3 cells decreases,the proliferation activity of the cells decreases(P <0.05),GH3 cells secrete GH(P <0.05)and IGF-1(P<0.05),and the percentage of apoptosis It was increased but not obvious.In the late stage of apoptosis,the proportion of apoptosis increased more obviously than in the early stage.Conclusion.The gene HS3ST2 may promote the occurrence and development of GH pituitary adenoma,and delay or reduce the apoptosis of tumor cells,and its main role node may be in the late stage of cell apoptosis.This study only confirmed some effects of the HS3ST2 gene on GH3 cells through in vitro experiments.We need to further explore the impact of its action pathways and animal experiments to study the role of the HS3ST2 gene in GH adenoma to show that the gene is GH pituitary gland Key genes for tumorigenesis and development.
