Objective:1.Establish the fever model of mice through "sticky disease"infection in Mongolian medicine under the guidance of Mongolian medicine theory.2.To establish the Heyi fever mouse model and its diagnostic criteria.Methods:Preliminary experiment:54 KM mice were randomly divided into low dose,medium dose,high dose group and blank group.There were 16 in the low,medium and high dose groups,and 6 in the blank group.The low,middle and high dose groups were anesthetized by intraperitoneal injection of 0.05ml 10%chloral hydrate,Fever model was established by intranasal instillation of Streptococcus pneumoniae with low dose(3×106CFU/ml,0.5?L/animal),middle dose(3×107CFU/ml,0.5 ?L/animal)and high dose(1.5×108CFU/ml,0.5 ?L/animal).The body temperature was measured every 2 hours after modeling,and the peak value,duration and symptoms of fever were observed.On the basis of the fever model of three dose groups,the"four-dose intervention" was given to the Mongolian medicine to promote the increase of "Heyi",including:behavioral intervention.The blank group was fed normally.normal feeding,summed up the symptoms of Hayi fever and established the diagnosis table of mice model of Hayi fever.Observe the change of symptoms,summarize the symptoms of Hayi fever and establish the diagnosis table of mice model of Hayi fever.The medium dose of streptococcus pneumoniae was determined as the experimental dose by observing the body temperature and the changes of symptoms.Formal experiment:1.Establish fever model of mice:intraperitoneal injection of 10%chloral hydrate(0.05 ml)shallow anesthesia Fever model group and Hayi fever model group,intranasal instillation of Streptococcus pneumoniae(3×107CFU/ml,0.5 ?L/animal),normal feeding of blank group.The blank group was fed normally.2.Establishment of Mongolian medicine Heyi fever model:On the basis of the fever of mice in the Heyi fever model group,the "four-dose intervention" was given to the Mongolian medicine to promote the increase of "Heyi",including:behavioral intervention(scare:70-80bB heavy metal noise),dietary intervention(strong tea:5g tea+400ml water boiled to the boil,placed to cool;Soba:usually the amount of diet 2/3,18g buckwheat flour and proper amount of normal temperature drinking water and to columnar),treatment intervention(tail vein bloodletting,every other day).The control group and the blank group were fed normally(adequate feed,drinking water)without special intervention.3.The contents of IL-1,IL-6,TNF-a,CRP,cTnT and so on in the serum of each group were determined by summarizing the symptoms of Heyi fever and the ELAISA method,and the changes in the tissue structure of the heart,lung and duodenum were observed under the electron microscope,To determine the model and diagnostic criteria of Hayi fever in mice.Results:After the model was built,the body temperature of the Hayi fever model group and the fever model group began to heat up at 11h and 10h,respectively,and reached a peak at about 20h.After 26h,the body temperature gradually decreased to the body temperature level of the blank group.In the process of fever,there were regular changes in spirit,activity,hair coat,respiratory frequency,water intake,urine color and odor,feces color and soft and hard degree,etc.,and the changes in symptoms during the fever period were consistent with the clinical symptoms of infectious diseases in Mongolian medicine.On the basis of the fever model of the Hayi fever model group,and the "four-dose intervention" that promoted the growth of "heyi" lasted for 7 days.The ears,nose and tail become lighter in color.The general state is sensitive,easily frightened,the activity increases,the amplitude increases;Insomnia;Drinking less water;Clear urine color,light smell;Excrement quantity is little,shape decrescent,quality of a material is hard,odour is weak wait for expression.These changes of symptoms are consistent with the symptoms of Heyi fever described by the Mongolian medicine theory of temperature disease and the symptoms in the diagnosis table of Heyi fever mouse model.Serum Il-1?Il-6?TNF-a?CRP and cTnT levels:Compared with the blank group,the content of TNF-a in fever model group and the content of TNF-a?IL-6?CRP?cTnT in the Hayi fever model was significantly different(P<0.05).Compared with the Fever model group,the content of IL-6 and cTnT in the Hayi fever model was significantly different(P<0.05).Conclusion:1.The nasal drip molding method of streptococcus pneumoniae is easy to operate in the experimental operation,the fever response is uniform,the peak of high fever is suitable,and the duration of fever is long,which is suitable for the establishment of heyi fever mouse model.2.The Mongolian medical theory promoted the intervention of Heyi's prosperity,which could increase Heyi in the course of fever and lead to Heyi's fever.3.It can provide a reasonable model for the pathogenesis of Mongolian febrile disease and related Mongolian medicine research.
