Background and Objective:Alzheimer's disease(AD)is the most common type of age-related dementia.According to age of onset,Alzheimer's disease is divided into early-onset(EOAD)and late-onset AD(LOAD)which is much more common.At present,the exact etiology and pathogenesis of AD are still being explored.AD is a complex disease with multiple factors,including genetic factors,environmental factors and immune factors.Currently,the ?4 allele of the apolipoprotein E(APOE)gene is the major risk factor for AD.And,the ?4 allele has been confirmed as playing a important role in AD.Although Genome-wide association studies(GWAS)and epigenome-wide association studies(EWAS)have identified some genes including APOE gene,most of the genes that contribute to LOAD is unknown.So,the other risk genes contribute to LOAD should be further explored.At present,some researchers have been shown the correlation between ANK1 and AD in epigenetic;on the other hand,GWAS data provide the evidence that the ANK1 locus is a novel common susceptibility locus for type 2 diabetes.AD and type 2 diabetes patients may have identical risk genes,this paper aims to explore whether the polymorphism of ANK1 rs515071 in Han population in northern China is associated with LOAD.Some researchers found AD and type 2 diabetes have the same risk allele or single-nucleotide polymorphism(SNP).Therefore,the aim of this study was to explore that the influences ANK1 gene single-nucleotide polymorphism in the risk of LOAD in a North Chinese Han populationMethods:We recruited a case-control study to investigate 900 cases and 1200 healthy controls matched for age and gender.All the participants were North Chinese Han population.SNP genotyping was performed using a SNP scanTM Kit(Cat#:G0104K,Genesky.Biotechnologies Inc.Shanghai)and APOE epsilon 4(APOE?4)genotyping was using improved multiplex ligase detection reaction(iMLDR).R 3.6.0 was applied for statistical.Wilcoxon rank sum test,chi-square test and Logistic regression test were used to this study.Result:In the APOE epsilon 4 carriers subgroup,the genotype distribution(p=0.033)and allele frequency(p=0.040)of rs515071 showed significant difference.In APOE?4 non-carriers subgroup,there is no significant difference.After adjusting for gender,age and APOE?4,we found that the mirror allele(A)increased the risk of LOAD in dominant model.Conclusions:In this study,we confirmed the ANK1 gene polymorphism is associated with Late-onset Alzheimer's disease in a north Han Chinese population.And,the mirror allele(A)increased the risk of LOAD by dominant model.
