Objective:The purpose of this paper is to explore the relationship between the endothelial microparticles proteomics screening differentially expressed proteins KRT8 and ANK1 in patients with coronary heart disease.Methods (1) Endothelial cells were cultured in vitro, using different concentrations of ox-LDL, TNF-stimulated endothelial cells. The use of real-time PCR technology to detect the expression of KRT8 and ANK1 inendothelial cells, and then, analysis of KRT8, ANK1 relative expression. (2) all recruited subjects underwent coronary angiography to select 50 patients confirmed by coronary angiography in patients with coronary heart disease,20 cases of patients with negative angiography as control group. We performed enzyme immunosorbent experiment to test the concentration of krt8 and ankl from the peripheral blood plasma of subjects. The degree of coronary stenosis in patients with coronary artery disease was scored using the standard of Gensini score, then the packet.Results (1) The stimulation of Ox-LDL HUVEC s can make the skeleton protein ANK.1, KRT8 expression, and different concentrations of ox-LDL stimulation can cause expression of HUVEC s ANK1, KRT8mRNA of cytoskeletal proteins in different; different concentrations of TNF-α stimulated HUVEC s could make the skeleton protein ANK1,KRT8 expression, and different concentrations of TNF-a stimulation, can cause expression of HUVEC s ANK1, KRT8mRNA of cytoskeletal proteins in different; (2) concentrations of KRT8 and ANK1 in patients with coronary heart disease was significantly higher than those without coronary heart disease group, P<0.05; (3) in patients with coronary heart disease subgroups, analysis between high achievers and low achievers in group KRT8, ANK1 concentration differences between the scores of Gensini, the P value of less than 0.05, there was significant difference between the groups, and ANK1,KRT8 concentration difference and Gensini score had positive correlation.Conclusion1.The stimulation of ox-LDL、TNF-α on HUVECs can increased the relative expression KRT8 and ANK1,in a concentration dependent manner.2. There is a certain correlation between KRT8, ANKl and coronary heart disease and the severity of the lesion, and has certain positive correlation.
