The main pathological changes in Osteoarthritis(OA)are the decrease or loss of articular cartilage matrix.In addition,synovial tissue hyperplasia,aseptic inflammation and other pathological changes may occur in Osteoarthritis.At present,the pathogenesis of osteoarthritis has not been studied.The clinical disability rate of osteoarthritis is very high,especially in the late stages of osteoarthritis of the knee,and there are often only a few single treatments.However,surgical treatment has the disadvantages of high cost and economic pressure.In terms of the diagnosis of osteoarthritis,the current clinical diagnosis of osteoarthritis can only be judged by the imaging data and the special features of the patient.Therefore,in order to improve the quality of life of osteoarthritis patients,reduce the incidence of osteoarthritis,and the discovery of new diagnostic markers is very important.In this way,patients with osteoarthritis can be diagnosed early,and then early intervention,early treatment.At present,many scholars are interested in the basic research of osteoarthritis.In general,seven Sirtuin proteins are present in mammalian organisms.Currently,a high acceptance rate holds that SIRT1 plays a role in extending cell life,fighting cell stress and promoting metabolism in mammals or lower organisms.SIRT1 is often associated with various diseases of aging,such as hypertension,diabetes,neurodegeneration,and joint degeneration.Therefore,in order to find genes that are effective in the diagnosis and treatment of some diseases related to aging,we hope to conduct further research on SIRT1 factor.According to relevant studies,SIRT1 plays an extremely important role in signaling pathways such as metabolism and inhibition of cell apoptosis in the body,mainly through deacetylation of lysine residues of transcription factors to regulate the high or low expression of relevant genes.According to the available information,SIRT1 is affected by a variety of other factors,such as the availability of substrates,the regulation of transcription factors and the expression of RNA proteins.In addition,the activity of SIRT1 is also related to its phosphorylation and methylation.Most importantly,the close relationship between SIRT1 and gene regulatory pathways of various transcription factors affects the expansion and differentiation of stem cells,which also affects the activity of SIRT1.In summary,by detecting the expression of SIRT1 in the cartilage tissue and serum of knee joint osteoarthritis in rabbits,this study aims to provide some new ideas for the clinical diagnosis and treatment of osteoarthritis,and ultimately improve the quality of life of patients with osteoarthritis.Objective:The expression of silencing information regulator 1 in the cartilage tissue and serum of the knee joint of experimental rabbits with different degrees of degeneration was analyzed to provide some new ideas for the gene therapy of osteoarthritis in the future.Methods:40 experimental white rabbits were randomly numbered and divided into 4 groups(group a,group b,group c and group d)according to the random number method,with 10 rabbits in each group.Group d was the control group,without any treatment.The knee osteoarthritis model of rabbits in group a,group b and group c was established by the method of straight plaster fixation.Group a was fixed for 6 weeks,group b was fixed for 8 weeks,and group c was fixed for 10 weeks.Before knee joint modeling in each group,adequate amount of venous blood was collected by ear vein method and placed in a test tube.After centrifugation for 10 minutes(3000 RPM),the upper serum was placed in a cryopreserved tube and frozen in a-20? refrigerator for inspection.After successful modeling,the serum samples of experimental rabbits were also collected by the same blood collection method and the same serum specimen preparation method.The experimental rabbits were sacrificed by air embolization,and the distal femoral cartilage tissue was taken and sectioned consecutively.The adjacent two groups were sectioned,one group was stained with HE,and the other group was examined by immunohistochemistry.After HE staining,Mankin score was performed according to the results.The cartilage sections were divided into 10 pieces of normal cartilage specimens,9 pieces of the mild degeneration group of osteoarthritis cartilage,10 pieces of the moderate degeneration group of osteoarthritis cartilage,and 9 pieces of the severe degeneration group of osteoarthritis cartilage.The adjacent 38 sections were grouped by immunohistochemical quantitative method and immunohistochemical detection of SIRT1 was performed to analyze the expression of SIRT1 in articular cartilage of rabbits in each group.Finally,the serum levels of SIRT1 were determined by enzyme-linked immunosorbent assay(ELISA).The expression of SIRT1 in serum of rabbits in each group was analyzed.Statistical analysis was conducted using IBM SPSS Statistics 21.0 software,and P<0.05 was considered statistically significant.Results:The results showed that the average gray value of SIRT1 in the normal cartilage group was 144.40±1.78,the average gray value of SIRT1 in the mild osteoarthritis group was 123.44±4.03,the average gray value of SIRT1 in the moderate osteoarthritis group was 112.50±4.97,and the average gray value of SIRT1 in the severe osteoarthritis group was 94.22±3.27.The gray value of SIRT1 in cartilage tissue decreased gradually with the degeneration of cartilage tissue.Through the building before and after building groups of experimental rabbit SIRT1 factor in peripheral blood of qing density is found:after building SIRT1 concentration(6.84+1.71)in serum is made before the mold(10.70±0.96)decreased obviously,has significant statistical significance(P<0.05),and further to build model before and after each group between experimental rabbit group statistical analysis found that:after building a,b,c groups of experimental rabbit serum concentrations of SIRT1 than concentration in front of the building is reduced,the statistical results are statistically significant(P<0.05).Conclusion:SIRT1 may play a key role in cartilage degeneration in osteoarthritis.The expression and concentration of SIRT1 in serum and cartilage tissue in cartilage tissues with more severe cartilage degeneration of knee joint were lower than that in normal cartilage experimental rabbits.
