Objective: to observe the effects of GSH on SOD activity,MDA content,il-6 and TNF-leupline expression in mice with acute kidney injury induced by agkistrodon halys venom,and to explore the protective mechanism of GSH on agkistrodon halys venom induced acute kidney injury in mice with agkistrodon halys venom.Methods: 60 female KMmice,according to the principle of randomized into 6 groups: normal control group(group A)reduced glutathione in the control group(group B)?low doses of snake venom intervention group(group C)? low doses of snake venom intervention + reduced glutathione in the treatment group(group D)?high doses of snake venom intervention group(group E)? high dose of venom intervention + reduced glutathione in the treatment group(group F)? With physiological saline respectively dealing with the normal control group(group A)and reduced glutathione in the control group(group B).Low dose snake venom intervention group(group C)and low dose snake venom intervention + reduced glutathione treatment group(group D)are injected with viper venom at 0.75mg/kg concentration.High dose snake venom intervention group(group E)and high dose snake venom intervention + reduced glutathione treatment group(group F)are injected with viper venom at 1.00mg/kg concentration.Three hours later,reduced glutathione(1800mg/kg)are injected into the snake venom intervention + reduced glutathione treatment group(group D and group F),and the same dose of reduced glutathione are injected into the glutathione control group.5 hours after the injection of reduced glutathione,blood and kidney specimens are collected,kidney pathological sections are prepared and scored.Serum creatinine(Scr),urea nitrogen(BUN),superoxide dismutase(SOD),malondialdehyde(MDA)are detected in peripheral blood,and interleukin-6(IL-6)and tumor necrosis factor-upe(tnfupe)were measured by ELISA.Results:1,The mice in group A and group B showed no abnormality,while the mice in group C,group E,group D and group F showed signs of lethargy,curled up,decreased activity,disorderly mouse hair,refusal of water and food after injecting viper venom.After the injection of reduced glutathione,mice in group D showed improved mental activity,increased feeding and activity,and showed no significant difference from group A and group B.Group F had occasional activities and eating,which is better than group E,but still has curled up and messy hair,which is still worse than group A or group B.2.SCr,BUN,MDA,SOD,TNF-? and IL-6 in group C,group E and group F are significantly different from those in group A(P<0.05)and group,SCr,BUN,MDA and TNF-? in group D showed no significant difference compared with group A(P>,0.05),SOD and IL-6 in group D are between group A and group C,and there are differences between the two groups(P<0.05).The differences of SCr,BUN,SOD,MDA,IL-6 and TNF-? in group F are significant compared with group A(P<0.05),but not statistically significant compared with group E(P>0.05).3.The renal tubular epithelial cells under pathological sections in group C and group E are swollen,the nuclei are shed,the tubular lumens are atrophic or collapsed,and some of the tubular lumens are protein-tubular,and a large number of vacuoles are observed between the cells.Interstitial infiltration of inflammatory cells(mononuclear or neutrophils)be observed,and interstitial dilatation is accompanied by tubule distortion,especially in group E and above,with significant reduction in group D,and no significant improvement in group F and group E.Conclusions:1.Reduced glutathione reduced MDA content,TNF-?,IL-6 expression and SOD activity in agkistrodon viper mice with acute kidney injury.2.The protective effect of reduced glutathione on kidney injury induced by agkistrodon halys venom in mice is related to the reduction of oxidative stress and inflammatory response...
