BackgroundAcute myocardial infarction(AMI)caused by coronary artery occlusion is one of the main causes of morbidity and mortality worldwide.Interventional therapy(intravascular balloon angioplasty and stent implantation)and surgical coronary artery bypass grafting are effective methods to restore blood flow,relieve angina pectoris,improve quality of life and reduce the risk of death from coronary heart disease,but MIRI is a common complication after blood flow recanalization.With the widespread development of coronary artery bypass grafting(CABG),ischemia-reperfusion(I/R)injury is inevitable during cardiac surgery.Therefore,in recent years,more and more attention has been paid to the study of MIRI.ObjectivesTo observe the effect of sevoflurane post-treatment on the levels of serum troponin(cTnI)creatine kinase isoenzyme(CK-MB)and lactate dehydrogenase(LDH)in(MIRI)model of myocardial ischemia-reperfusion injury in rats.To investigate whether sevoflurane post-treatment has protective effect on myocardial ischemia-reperfusion injury in rats.MethodsThirty healthy adult male SD rats weighing 250g-300 g were randomly divided into three groups: control group(Group C),ischemia-reperfusion group(Group R),sevoflurane post-treatment group(Group S).Myocardial ischemiareperfusion model was established by ligation of LAD.In the control group(Group C),the silk thread only passed through the lad without blocking the blood flow for 150 minutes;in the ischemia-reperfusion group(Group R),the blood flow was blocked for 30 minutes and then reperfusion for 120 minutes;in the sevoflurane post-treatment group(Group s),2% sevoflurane was inhaled immediately for 15 minutes during the reperfusion period after 30 minutes,the rest of which was the same as that in the R group.The heart rate(HR)and mean arterial pressure(map)of the three groups were recorded during open chest basal value(T0),ischemia for 10min(T1),ischemia for 30min(T2),and reperfusion for 30min(T3),60min(T4),120min(T5).The changes of serum troponin I(cTnI),creatine kinase isoenzyme(CK-MB)and lactate dehydrogenase(LDH)were measured after reperfusion.The rats were killed by air embolism after the experiment.Results1.The model of myocardial ischemia-reperfusion injury was successfully established by ligating the left coronary artery2.Observe the hemodynamic changes of the three groups at different time points.It is found that there is little difference in heart rate changes of the three groups at T0,T1 and T2.At T3,T4 and T5,there is no significant difference in heart rate changes of group C.compared with Group R,s and C,the heart rate decreases significantly(P < 0.05),but there is no significant difference between the two groups(P > 0.05).The blood pressure of the three groups is at T0,T1,T2,There was no significant change between the time groups,but with the time going on,the blood pressure had a downward trend.With the time going on,the blood pressure of Group R and group s was significantly lower than that of group C(P < 0.05),and that of Group R and group s was significantly lower than that of group C(P < 0.05).3.The levels of cTnI,CK-MB and LDH in the three groups were significantly higher than those in group C(P < 0.05)and Compared with S group,R group was higher than S group(P < 0.05).Conclusion1.Sevoflurane post-treatment can improve the blood supply of myocardial ischemia-reperfusion injury.2.Sevoflurane post-treatment reduced the release of myocardial enzymes,improved myocardial injury after reperfusion,and had a certain protective effect on myocardial ischemia-reperfusion injury.
