Abstract/Summary:
Objective:To investigate the relationship between glycoprotein synthase kinase-3(GSK-3β)and mitochondrial cleavage protein,Drp-1,in the restoration of protective effect of sevoflurane post-treatment on myocardium in diabetic rats.Methods:sixty healthy male Sprague-Dawley rats(250~300 g),were given adaptive feed for 1 week.A diabetic rat model was prepared by intraperitoneal injection of streptozotocin(STZ)30 mg/kg after fasting for 12 h.The successful SD models were randomly divided into 5 groups(n=8 each)using a random number table:Ischemia reperfusion group(group I/R),seven sevoflurane postconditioning+Ischemia reperfusion group(group SP).Ischemia reperfusion+sevoflurane postconditioning+Mivi-1 treatment group(group SM),Ischemia reperfusion+sevoflurane postconditioning+SB216763 treatment group(group SM),and Ischemia reperfusion+sevoflurane postconditioning+Mivi-1 treatment group+SB216763 treatment group(group SS).The myocardial ischemia-reperfusion injury model was established by ligating the left anterior descending coronary artery for 30 min and reperfusion for 120 min.Group SP,SM,SB and group SS were given sevoflurane at 5 min before reperfusion,and the end-expI/Ratory concentration was 2.5%,lasted for 10 mins.DMivi-1(1.2 mg/kg)was injected intravenously 15 min before reperfusion in group SM.SB216763(0.2 mg/kg)was injected intravenously 5 min before reperfusion in group SB.Both of them were injected intravenously 1 min before reperfusion in group SS.Serum ardiac troponin I(cTn I)concentration was detected from abdominal aorta blood.Myocardial specimens were obtained from the apex for determination of the cell apoptosisand apoptotic index(AI)by TUNEL.Expression of caspase-3was detected by Western blot Myocardial pathology were observed and the myocardial pathology were observed.Results:Compared with I/R group,the levels of caspase-3,AI and myocardial injury in SP group were not statistically significant(P>0.05).Compared with I/R group,SM,SB and SS groups were down-regulated(P<0.05).Compared with SP group,the indicators above in SM,SB and SS groups were down-regulated(P<0.05).Compared with SS group,the levels of caspase-3,AI and myocardial injury were increased in SM and SB group(P<0.05).Conclusion:The relationship between GSK-3β and Drp-1 in the restoration of protective effect of sevoflurane post-treatment on myocardium in diabetic rats is not a single dependency.
