A Metabolomics Study Of Traditional Chinese Medicine Recipe "Qing Luo Yin" In Treating Heat-Induced Paralysis Syndrome

Objective: Qing Luo Yin is a traditional Chinese medicine master's experience in treating damp heat syndrome,and its treatment mechanism is unclear.This study used metabolomics to initially explore the material basis and affected metabolic pathways of Qingluo Yin for damp heat syndrome.Research lays the foundation to better guide clinical medication.Methods: Collect the blood and urine of the patients who used qingluoyin in the treatment of Bi syndrome in damp heat period before treatment,for one month,for two months and for three months.Detect the metabolites in the blood by the method of non target metabolomics,and analyze the single variable and multivariate analysis to find the different metabolites before and after treatment,and analyze the metabolic pathway.Results: Through the detection and analysis of metabolites in blood and urine,there were 71 metabolite multiple changes(FC)greater than 2 and Student t in blood after one month of continuous administration p value was less than 0.05,but FC value of 179 metabolites was more than 2 and p value was less than 0.05 after two months of continuous administration,FC value of 603 metabolites was more than 2 and p value was less than 0.05 after three months of continuous administration.There were 51 metabolites in urine with FC greater than 2 and p value less than 0.05 at one month,but 172 metabolites with FC greater than 2 and p value less than 0.05 at two months after continuous administration,and 72 metabolites with FC greater than 2 and p value less than 0.05 at three months after continuous administration.The primary metabolites were identified by orthogonal partial least square method.The main metabolites of blood difference were carbamoyl phosphate,phenylalanine,diacylglycerol,coenzyme A,diacylglycerol,phosphatidylic acid,phosphatidylcholine,phosphatidylserine,phosphatidylethanolamine,lysophosphatidyl,cardiolipin,L-acetylcarnitine,bile acid,glycine,serine,homotype Hemi The contents of cystine,adenine,phosphatidylglycerol and other metabolites in urine are mainly creatine,deoxycreatinine,diacylglycerol,L-phenylalanine,1,Contents of 7-dimethylguanosine,inositol phosphate,hydroxyprolinyl lysine,pantothenic acid,linoleic coenzyme A,flavin mononucleotide,glyceryl diester,phosphatidylglycidyl phosphate,guanine,lysine,diacylglycerol,phosphatidylethanolamine and other metabolites.These changes are mainly related to lipid metabolism,amino acid metabolism,purine metabolism,pyrimidine metabolism and oxidation-reduction pathway.Conclusion: With continuous Qing Luo Yin treatment,patients with damp-heat paralysis have undergone significant changes in body metabolism.At different time periods after continuous medication,the metabolite changes in patients are not all the same,but have similar characteristics.Metabolites related to synthetic raw materials or metabolites of amino acids and lipids in patients' serum have changed,and metabolites related to synthetic raw materials or metabolites of amino acids,lipids and purines have changed in urine.These metabolites can play an important role in pain relief,energy substrate supply and muscle recovery,muscle cell proliferation,protein homeostasis regulation,tissue response to inflammation,cellular oxidative stress,and repair of injured joints.By analyzing metabolic pathways of metabolites that have undergone significant changes,the metabolic pathways affected by Qingluo Drink are mainly related to the synthesis and metabolism of amino acids,energy,lipids,purines,etc.Lipid metabolism affects wound healing and the severity of arthritis after joint injury.Purine-related metabolic pathways affect urate content,related media,and alleviate pain in patients.Amino acid and energy metabolism pathways affect various mechanisms such as oxidative stress,enhanced stress resistance and recovery ability.
