| Chinese agarwood is the aromatic resinous heartwood obtained from Aquilaria sinensis (Lour.) Gilg, and is famous and precious traditional Chinese medicine. Sesquiterpenes and2-(2-phenylethyl)chromone derivatives are the main and characteristic components of agarwood. And2-(2-phenylethyl)chromone derivatives include5,6-epoxy-2-(2-phenylethyl)chromones,5,6:7.8-diepoxy-2-(2-phenylethyl)chromones,5,6,7,8-tetrahydro-2-(2-phenylethyl)chromones and flidersia type2-(2-phenylethyl) chromones. The2010edition of "Pharmacopoeia of people’s Republic of China" records that morphological identification, color identification, determination of extractives, microscopic identification and TLC identification for identification and quality evaluation of agarwood.In order to provide more scientific quality evaluation methods of agarwood, four kinds of qualitative methods and a quantitative method were used to analyze a ten batche of Chinese agarwood samples. The method of TLC can analyze the common and characteristic color spots of agarwood easily and quickly. The method of GC-MS can qualitative identify the volatile sesquiterpenes and some flidersia type2-(2-phenylethyl)chromones of agarwood. The method of HPLC and HPLC-MS can be qualitative analysis of the four common types of chromones of agarwood. And UV can determine the absolute content of all2-(2-phenylethyl)chromones of agarwood rapidly.The improved TLC method which include two kinds of deployment systems and three kinds of color methods, were used to analyse the ten samples. The results were that seventeen common color spots were found and six of them were characterized by eight standard compounds. The TLC fingerprint method was established, and it can use to identify the authenticity of a sample by testing the presence or absence of the common characteristic color spots of agarwood.The total ion chartes of ten batches of agarwoods were got by GC-MS, and GC-MS fingerprint method was established. A total of ninety chromatographic peaks were detected, seventy-five of them was identified, of which eleven identified compounds were common. The total ion charte was divided in sesquiterpenes area and2-(2-phenylethyl) chromones area based on the qualitative analysis and the retention time of chromatographic peaks. Through the qualitative analysis of a sample mainly with characteristic of sesquiterpenes and chromone compositions of agarwood to identify the authenticity.The HPLC was used to analyse the agarwoods. The result was that fingerprint method was built, and the chromatographic peak could be characterized by the retention time of standard compounds. The result of similarity evaluation showed that similarity higher than0.80of the samples accounted for90%. The result illustrated that the similarities of the compounds in the samples were relatively high, but there were still some differences.Twenty-four standard compounds of2-(2-phenylethyl)chromones was analysed by HPLC-MS. The MS identified method of four kinds of chromones was summarized. Seventy-six compounds were detected by HPLC-MS from the ten samples, including15common components. Through the analysis of relative amount of chromones to identify the authenticity of samples. And the relative formation time often agarwoods can judge on the basis of reported biogenetic pathway of four kinds of chromones.According to GC-MS qualitative result, ten samples were divided into three categories. The samples were divided into five categories on the basis of the HPLC-MS qualitative result. The cluster results were differences, mainly because of the GC-MS clustering method was based on the common volatile components, and the HPLC-MS clustering method was based on the common2-(2-phenylethyl)chromones.The four kinds of qualitative methods were used to identify two samples. The four methods of analysis draw the same inference. The first sample was the low-quality agarwood with other constituents, and the other sample was other plant which add some extracts of agarwood.After the authentication of the agarwood, UV method was established for determination of the absolute content of2-(2-phenylethyl)chromones in agarwood. According to their chromones percentage content, ten samples were divided into three groups.The establishment of the five kinds of analysis methods can provided more scientific identification method for quality evaluation and scientific evidence for the establishment of quality standard of agarwood. |
