Objective To investigate the mechanism of polygonum cuspidatum alcoholic extract interfering with acute gouty arthritis induced by MSU through NLRP3/ASC/Caspase-1 axis.Methods ?Using the total flavonoids of Polygonum cuspidatum as indicator,the extraction technology of ultrasonic extraction of total flavonoids from Polygonum cuspidatum was investigated by orthogonal test.?The effects of different factors on the preparation of MSU injection suspensions were observed by microscope.?The model of acute gouty arthritis mice was constructed by different modeling methods,and the modeling method was determined by the index of the volume swelling rate,inflammatory index grading,gait scoring,observation of the pathological changes of the ankle synovial membrane and the determination of i1-1? content by Elisa.? Using Polygonum cuspidatum alcohol extract High school three doses,with colchicine as a positive control drug,in the sixth day of irrigation with MSU model,the seventh day after the last time to take samples of medicine,Using the total flavonoids of Polygonum cuspidatum as alcohol,the extraction technology of ultrasonic extraction of total flavonoids from Polygonum cuspidatum was investigated by orthogonal test.ELISA was applied to determine the changes of ankle joint fluid IL-1??IL-6 and TNF-a.Testing the expression of protein NLRP3?ASC and Caspase-1 by Western blotting.The expression of NLRP3?ASC and Caspase-1 gene were tested by RT-PCR.Results The optimum extraction process of total flavonoids of 1 Polygonum cuspidatum was as follows:Ethanol quality fraction 65%,material liquid ratio 1:40,ultrasonic extraction of min,the total flavonoids extraction rate was 19.69%under the extraction condition.MSU uses 0.9?sodium chloride as a solvent and uses a vortex to promote the dispersion of 50g/L concentration.This concentration is not easy to block the needles used in the experimental model.The model of acute gouty arthritis mice was made by ankle joint model,and ankle joint fluid and ankle synovial membrane were taken as the sample.Through the analysis of the result of the volume swelling rate of the toe in mice,the effect of high dosage group of ethanol extracts from Polygonum cuspidatum on the intervention of acute gouty arthritis mice induced by MSU in the lower dosage group was obvious,which inhibited the expression of IL-1?,IL-6 and TNF-a,and NLRP3 and ASC in the synovial tissue of ankle joint,Caspase-1 protein and mRNA play an intervening inhibition.Conclusion From the animal level,the Polygonum cuspidatum may interfere with acute gouty arthritis by inhibiting the expression of IL-1?,IL-6 and TNF-a,synovial tissue NLRP3,ASC,Caspase-1 protein and mRNA.Provides a basis for further exploration and research into the related material of the Polygonum cuspidatum,which is effective in the future.
