Jiuxiangchong,the traditional Chinese medicine,is the dry body of Aspogopus chinensis Dallas,which has the effect of regulating qi to alleviate pain and warning splenic Yang recipes.It is used for cold and distending pain of stomach,stomachache due to emotional depression and the hyperactive liver Qi attacking the stomach,impotence due to kidney deficiency to stomach weakness of the kidney,aching lumbus and knees.At present,there are more and more mixed use and misuse of Aspogopus in the market,and its quality problem has attracted much attention.It is of great significance to standardize the market management of Aspogopus,improve its quality evaluation system and ensure its quality.Objective: To investigate and identify Aspogopus and its adulterants,formulate grade standard for commodity specification,and establish a systematic identification system combining character,microscope,physicochemical and molecular identification methods,so as to provide reference for the accurate identification and quality control.Methods:(1)Through the origin and market surveys,and specimen collection,grasp the resource distribution of the Aspogopus.In addition,the collected Aspogopus medicinal materials were measured and analyzed according to the appearancecharacteristics,and established the standard of commodity specifications and grades of Aspogopus.(2)The establishment of a basic identification system for Aspogopus use animal taxonomy,medicinal material traits,genital identification,micro identification,micro identification,thin layer chromatography method,etc.(3)Based on the DNA barcode and mitochondrial whole gene sequencing method to establish the molecular identification system of Aspogopus.(4)Analyze the composition of fatty oils based on GC-MS technology,analyze the polysaccharide composition of Aspogopus based on HPLC-derivative method,and explore the identification system of Aspogopus based on ingredients.Results:(1)Accumulatively surveyed and identified 8 species of Aspogopus and its adulterants,all of which belonged to Pentatomidae family with similar traits.And initially established the Chinese medicine Aspogopus product specification grade standard.(2)Summarized the trait characteristics,genital characteristics,and micro-traits and micro-characteristics based on the ventral and back plate as the main identification basis for Aspogopus and its adulterants.(3)The results of DNA barcode identification showed that all varieties except Aspongopus nigriventris westwood can be distinguished well;the whole mitochondrial gene sequencing method found that the Aspogopus consisted of two mitochondrial genes that differ greatly.(4)The composition of fatty oils of Aspogopus and its adulterants are generally similar by GC-MS technology.Through fingerprint similarity analysis and systematic clustering analysis,Aspogopus and its adulterants are difficult to distinguish,except Tessaratoma papillosa Drury.An analysis of its polysaccharidecomposition found that the polysaccharide content of Cyclopelta Parva Distant was relatively high,followed by Megymenum inerme H.S;The polysaccharides of Aspogopus and other adulterants are mainly composed of mannose,rhamnose,glucose,galactose,etc.,while the polysaccharides of Cyclopelta Parva Distant are mainly composed of glucose;Judging from the principal component analysis,except for Aspongopus nigriventris westwood,other varieties can be aggregated into one category,which can be better distinguished from Aspogopus.Conclusion: Based on the systematic identification of Aspongopus,it can better distinguish other adulterants except for Aspongopus nigriventris westwood.Aspogopus may have hidden biodiversity.Using the method of analyzing the composition of polysaccharides,it can be used for the identification of the varieties of Aspogopus.
