Ganoderma atrum has been used for more than two thousand years in Asian countries.Modern medicine also confirmed that Ganoderma atrum have many biological activities such as enhancing immunity,suppressing tumor growth,reducing blood-lipid,decreasing blood pressure and resisting oxidation.Polysaccharide is regarded as the major bioactive component in Ganoderma atrum.The present study is designed to explore the anti-inflammatory effect of PSG-1,which is isolated and purified from Ganoderma atrum cultivated in Ganzhou,Jiangxi province,using cell culture and molecular biology technologies in vitro.This study based on LPS-induced inflammatory macrophages model and intestinal epithelial cells/macrophages co-culture inflammation model,in order to illustrate the anti-inflammatory effect and potential mechanism.The main research results of this study are concluded as follows:1.PSG-1 has a protective effect on LPS-induced inflammatory macrophages model,which may be related to reducing the expression of proinflammatory cytokines(TNF-?,IL-6,IL-1?),COX-2,ROS and other inflammatory mediators to resist LPS induced inflammatory response,down-regulating the phosphorylation level of MAPKs pathway and activating Nrf2 pathway.2.LPS-induced Caco-2-RAW 264.7 co-culture intestinal inflammation model was established to simulate the intestinal microenvironment.The results indicated that PSG-1 reduced the permeability of Caco-2-RAW 264.7 co-culture model by increasing the activity of LAP and DAO,as well as up-regulating the expression of three tight junction proteins of ZO-1,Occludin and Claudin-1,thereby improving LPS stimulation Impaired barrier function.3.PSG-1 has a protective effect on LPS-induced Caco-2-RAW 264.7 co-culture model.In addition,PSG-1 not only has a direct anti-inflammatory effect on LPS-induced macrophages,but also has an indirect anti-inflammatory effect on macrophages under the barrier of intestinal epithelial cells.And it can inhibit the overexpression of proinflammatory cytokines(TNF-?,IL-6,IL-1?),COX-2,ROS and other inflammatory mediators,regulate the phosphorylation level of the MAPKs pathway and activate the Nrf2 pathway,to play an anti-inflammatory role.
