Objective: Dangguiliuhuang decoction is a classic prescription in the secret collection of the orchid chamber compiled by Li Dongyuan in the Jin Dynasty.In this paper,we will establish the preparation process of the traditional material standard of Dangguiliuhuang decoction,and carry out the experimental research on the quality attribute of the material standard of Dangguiliuhuang decoction,and carry out the consistency evaluation and the research on the quantity and quality transfer rule of the material standard of Dangguiliuhuang decoction one step development research provides theoretical basis.Methods: Through consulting a large number of ancient medical books and literature,the prescription source,prescription dose,usage and dosage,efficacy and main treatment of Dangguiliuhuang decoction,as well as the prescription of traditional Chinese medicine,drug location,preparation process and other contents were examined;according to the 2015 edition of Chinese Pharmacopoeia,the quality of collected batches of medicinal materials was evaluated,and high-quality medicinal materials were selected to prepare pieces for preparation Feeding;following the records of ancient books,combining with modern practical operation,determining the preparation process of traditional decoction;drying the decoction by vacuum freeze-drying method to obtain the material standard of Dangguiliuhuang decoction;experimental research on the quality attribute of the material standard of Dangguiliuhuang decoction by thin layer chromatography and liquid chromatography,and then evaluating the consistency of the material standard of Dangguiliuhuang decoction Study on the law of quantity and quality transfer in the price and its medicinal materials-pieces-material standard.Results: Dangguiliuhuang decoction is a kind of decoction in the secret collection of the orchid chamber compiled by Li Dongyuan in Jin Dynasty.All the herbs used are the regulated sources of Pharmacopoeia.The preparation process is: smash 2.58 g of angelica,raw rehmannia,cooked rehmannia,Scutellaria,Coptis,Huangbai and 5.16 g of Astragalus into coarse powder,add 400 ml of water,decoct for about 20 min,to 200 ml of decoction,filter,freeze-drying,and get Dangguiliuhuang decoction qualitative benchmarks.The specific identification of angelica,Coptis,Scutellaria and Astragalus in the material standard was established by TLC;the content determination method of Baicalin in the material standard of Dangguiliuhuang decoction was established by HPLC.The results were in accordance with the regulations by the method of the law of the other side and the applicability of the system.The results showed that the method could be applied to the content determination of Baicalin in the material standard The HPLC fingerprint of Dangguiliuhuang decoction was established,and 15 batches of Dangguiliuhuang decoction were investigated,27 peaks were identified.The RSD values of the relative retention time and the relative peak area of each common peak are less than 3.0%,which shows that the method is stable and feasible.Through the consistency evaluation of 15 batches of Dangguiliuhuang decoction,the results showed that the average extraction rate was 42.24%,RSD was 2.74%;the average extract content was 73.41%,RSD was 3.01%;the similarity of fingerprints was greater than 0.98,and the consistency evaluation results were good;through the analysis of the quantity and quality transfer rule of Dangguiliuhuang decoction,the results showed that the average transfer rate of Baicalin in herbs pieces was 42.24% It was 95.79%;RSD was 2.75%,and the average transfer rate of baicalin was 57.97%;RSD was 7.86%;according to the study of fingerprint,the corresponding effective substances were passed down step by step,which indicated that the process was feasible and the relationship between quantity and quality was good.Conclusion: According to the ancient medical books and documents,the composition,dosage and the traditional preparation method of the prescription were determined,and the research method of the material standard quality attribute of Dangguiliuhuang decoction was established.The quality evaluation and quality control of the material standard of Dangguiliuhuang decoction were carried out by the methods of TLC identification,baicalin content determination and fingerprint The law of quantity and quality transfer of price and its medicinal materials-pieces-material standard provides a scientific theoretical basis for the further development and research of the classic prescription Dangguiliuhuang decoction.
