Objective: After abdominal massage stimulates the intestinal nervous system,the brain function can be improved through brain gut interaction,and then primary insomnia can be treated.The experimental object is the patients with primary insomnia,which involves medical ethical issues,and can not better reveal the mechanism of brain gut interaction.Through the design of the animal experiment of brain gut interaction mechanism,this study expounds the relationship between brain gut peptide and primary insomnia under the brain gut interaction mechanism,and provides theoretical basis for the treatment of primary insomnia with traditional Chinese medicine.Methods: In this study,the insomnia model of rats was reproduced by PCPA method.The animal model was evaluated by macroscopic performance and experimental indexes.The content of substance P,galanin,?-endorphin and collagen fibrillary acidic protein in serum,hypothalamus and small intestine of rats were determined by enzyme-linked immunosorbent assay,and the change of the content of brain gut peptide in insomnia rats was observed The effect and mechanism of massage on primary insomnia.Results:(1)After PCPA method,insomnia rats appeared,dry hair,restlessness at night and day,dry stool,increased water consumption and reduced diet.After pentobarbital sodium injection,the time of righting reflex was significantly prolonged,and the symptoms of massage group were improved after treatment.There was no significant difference in the serum GFAP content of the blank group on the 5th and 10 th day(P > 0.05).Compared with model group,massage group and drug group on the 5th and 10 th day,the serum GFAP content decreased(P < 0.05).There was no significant difference in serum GFAP content between the groups after 5-day treatment and 10-day treatment.(2)There was no statistical difference between the model group,the massage group and the drug group in the content of SP in the serum,the hypothalamus and the small intestine of the rats after 5 and 10 days.The SP content of serum,hypothalamus and small intestine in model group was lower than that in other groups after 5 days.The SP content of hypothalamus in model group was lower than that in other groups after 10 days.(3)There was no statistical difference in the serum,hypothalamus and small intestine of the blank group.The serum and hypothalamus gal content of the model group,massage group and drug group was higher than that of the five days later,and there was statistical difference.There was no significant difference in the content of gal in small intestine.Single factor analysis of variance was used to compare the gal content in serum,hypothalamus and small intestine of rats after 5 days.The gal content in serum and hypothalamus in model group was lower than that in other groups,with statistical difference.The gal content in small intestine in model group was higher than that in other groups,with statistical difference.The gal content in hypothalamus in model group was lower than that in other groups after 10 days,with statistical difference.There was no statistical difference in serum and small intestine.(4)The content of ?-endorphin in serum,hypothalamus and small intestine of rats at 5 and 10 days was compared by t-test between groups.There was no statistical difference in the blank group.The content of ?-endorphin in hypothalamus and small intestine of rats in model group,massage group and drug group after 10 days was higher than that after 5 days,and there was statistical difference.The content of serum ?-endorphin in massage group and drug group was higher than that in 5 days after 10 days.There was no statistical difference in the model group.The content of ?-endorphin in serum,hypothalamus and small intestine of rats was compared by one-way ANOVA after 5 days.The content of ?-endorphin in model group was lower than that in blank group,with statistical difference.The content of ?-endorphin in hypothalamus of model group was lower than that of other groups after 10 days,with statistical difference.There was no statistical difference in serum and small intestine.Conclusion:(1)PCPA method successfully reproduced the insomnia rat model,the serum GFAP content and the degree of sleep were negatively correlated,abdominal massage group and drug group can reduce the GFAP content,improve insomnia symptoms.(2)After treatment,the content of SP in serum,hypothalamus and small intestine increased,suggesting that it is positively related to the degree of sleep.Abdominal massage group and drug group can improve sleep by improving the content of SP in the body.(3)After treatment,the levels of serum and hypothalamic gal increased,suggesting that the levels of serum and hypothalamic gal were positively correlated with the degree of sleep,while the levels of small intestine gal decreased,suggesting that sleep was negatively correlated with the level of small intestine gal.Massage group and drug group can improve sleep by improving gal content in vivo.(4)After treatment,the content of ?-EP in serum,hypothalamus and small intestine increased,which indicated that the content of ?-EP was positively related to the degree of sleep.In conclusion,abdominal massage can improve insomnia symptoms by regulating brain gut peptide.
