Objective:In this study,a rat model of premature ovarian insufficiency(POI)induced by intraperitoneal injection of Cyclophosphamide(CTX)was used,and it was intervened by the use of "Zhibian Point to Shuidao Point" acupuncture acupuncture.Acupuncture therapy and western medicine therapy are compared.After treatment,the serum hormone levels and bcl-2 and bax protein levels in the ovary of each group were measured,and the therapeutic effect of the Zhibian percutaneous acupuncture method on POI rats was observed.Incompleteness provides more effective treatments,and at the same time expands the range of treatments for rank-edge percutaneous acupuncture.Methods:A total of 60 female SD rats with normal estrous cycle were selected,and their body weight(200 ± 20)g at eight weeks of age was randomly divided into blank group,model grouP,"Zhibian Point to Shuidao Point" group(rank side pervious channel),and ordinary acupuncture group(Shenshu,Guan Yuan,Sanyinjiao),western medicine group(estradiol),12 in each group.Except the blank group,the other four groups were injected intraperitoneally with cyclophosphamide for fifteen days,and the vaginal smear was taken to verify the success of the model ten days before the end of the model.The treatment was started the next day after model establishment.The general acupuncture group and the Zhibian group were treated with acupuncture every other day.The western medicine group was treated with daily estradiol.Fixed restraint,the treatment cycle is one month.The samples were collected uniformly the day after the end of the treatment.Blood was collected from the abdominal aorta,and the ovaries and uterus were collected.ELISA was used to detect the content of E2,FSH,LH,AMH,and INHB in serum,and the expression of bcl-2 and bax proteins in ovaries were detected by immunohistochemistry and immunoblotting,respectively.Follicles in ovaries were observed by HE stain Changes and changes in the uterus.Results:1.Comparison of general conditions: During the modeling period,compared with the blank group,all model rats had a significant decrease in body weight,increaseddrinking water,reduced diet,and appeared dull and dull,frizzy,kinky,uncomfortable,Partially large The stool was sparse and spontaneous,and healed occasionally after blood in the stool.Hair loss was apparent in the back of the neck and the top of some rats.The symptoms of the three treatment groups were significantly improved at the beginning of treatment.2.Comparison of serum hormone levels: Compared with the blank group,the levels of FSH and LH in the serum of the model group significantly increased,and the content of E2 decreased significantly.The difference was statistically significant(P < 0.05).Compared with the model group,the serum FSH and LH levels of the rats in the rank side group were significantly reduced and the E2 levels were significantly increased,and the difference was statistically significant(P<0.05).Compared with the blank group,the FSH,LH,and E2 contents in the serum of the rats in the rank side group were not significantly different(P>0.05).AMH and INHB,as indicators of ovarian reserve function,were significantly reduced in the model group.Except that there was no significant difference between the AMH and the model group,the levels of AMH and INHB in the serum of the other three groups were significantly different from those in the model group.Statistical significance(P<0.01).The levels of AMH and INHB in the serum of rats in the rank side group were significantly higher than those in the other treatment groups,and the difference was statistically significant(P<0.01).3.The expression of Bcl-2 and bax in the ovary: bcl-2: Compared with the blank group,the expression of bcl-2 Protein in the model group was significantly reduced,and the difference was statistically significant(P<0.05).Ratio,the protein expression in each treatment group increased to varying degrees,except for the general acupuncture group and the difference was not statistically significant,the bcl-2 protein content in the Chibian group and the Western medicine group was compared with it(P<0.05),The differences were statistically significant;compared with the rank side group,there was no statistically significant difference between the bcl-2 protein and the western medicine group(P>0.05).Bax: Compared with the blank group,the expression of bax protein in the model group was significantly increased,and the difference was statistically significant(P < 0.01).Compared with the model group,the expression of bax protein in each treatment group was reduced to varying degrees.The expression of bax protein in the rank side group,thegeneral needle group,and the western medicine group had significant differences(P <0.05).Compared with the rank side group,the protein expression in the blank group and the western medicine group had no significant statistics Significance(P> 0.05).4.Changes in ovarian tissue and follicles: Rats in the blank group had a full ovary shape and ruddy color.Follicles and corpus luteum at various levels were visible in the ovary,and the number of occluded follicles was small.Compared with the blank group,the ovaries in the model group had smaller volumes Dull gray hair,markedly increased atresia follicles in the ovary,almost no visible follicles during growth;the ovary volume and appearance of rats in the rank side group were not significantly different from the blank group,and the color was ruddy than the model group,but less bright than the blank group,and normal in the ovary The number of developing follicles and corpus luteum,vacuole follicles and atretic follicles were significantly reduced,and the ovarian structure was similar to the normal group.Conclusion:1.The pervious acupuncture method can effectively improve the ovarian structure and follicular quality of rats with early-onset ovarian insufficiency.2.Rank-edge permeable acupuncture can effectively regulate the hormone levels in the serum of rats with early-onset ovarian insufficiency,reduce the levels of FSH and LH in the serum,increase the levels of E2,AMH and INHB,and promote the restoration of the ecological balance of the HPOA axis.3.The regulation and treatment effect of the Zhibian permeable acupuncture method on rats with early-onset ovarian dysfunction may be achieved by inhibiting the apoptosis of granulosa cells in the ovary.This acupuncture method can effectively regulate bcl-2 and Bax protein expression.
