Objective: To investigate the distribution characteristics and medicinal materials of Crepis crocea in Shanyin County by taking the opportunity of the fourth national resource census,and to explore the chemical components and medicinal effects of Crepis crocea on the ground and roots.Yangshen,which is a special medicine in Shanxi,provides a scientific basis.Method:1 Investigation of wild resources of Shanyin Crepis crocea according to the nation’s fourth general survey plan of traditional Chinese medicine resources,field surveys and field sample surveys were used to conduct field investigations on the distribution and reserves of Crepis crocea resources in Shanyin County.The distribution characteristics and medicinal material reserves of Crepis crocearesources in this ecological area.2 Study on the chemical composition of different medicinal parts of Crepis crocea: high-performance liquid chromatography to determine the content of chlorogenic acid,ultraviolet spectrophotometry to determine the content of polysaccharides,and hot dip to determine the content of extracts;The NMR hydrogen spectrum of different parts of Crepis crocea was analyzed,and its chemical composition was identified,and the different compounds in different medicinal parts were screened,and the chemical composition of different parts of Crepis crocea was evaluated as a whole.3 Contrastive study on the efficacy of different medical parts of Crepis crocea: Using the concentrated ammonia solution of mice to induce cough,endotoxin-induced inflammation in rats,and cyclophosphamide-induced mouse immunosuppression model to study the efficacy of different medical parts of Crepis crocea Compare the possible differences in the efficacy of different parts.Results: 1 Investigation on the distribution and reserves of Crepis crocea resource.Shanyin County is located in the northern part of Shanxi Province,with an area of 1657 square kilometers.According to the census plan,it is divided into five representative areas of grassland,grassland,meadow,shrubland,and broad-leaved forest according to its vegetation type.A set of sample plots.Through investigation,it was found that Crepis crocea is distributed in four representative areas of grass,grassland,shrubland,and broad-leaved forest.The distribution area of Crepis crocea in Shanyin County is about 8.954Km2,and the medicinal material reserves are about 50.4189 t.Among them,the reserves of Crepis crocea medicinal materials in each ecological representative area from high to low are shrubs,grasslands,grasses,and broad-leaved forests.2 Chemical composition of different medicinal parts of Crepis croceaThe chlorogenic acid content of the ground part of Crepis crocea is close to 0.5%,and the chlorogenic acid content of the root is less than 0.095 ~ 0.333%;The polysaccharide content in the grass and aboveground parts,and also in the aboveground part of Huanyangshen is lower than 0.12 ~ 3.96%.The root extracts of Crepis crocea collected from June to July were all between 36.98 and 52.54%,and were higher than the above ground content.From August to September,the content of the above ground part of Crepis crocea increased,from 29.51 to 49.03 %,And higher than the root.The number of peaks in the UPLC map of Crepis crocea on the ground and roots differed greatly,and the content of the same component also differed greatly.There were 22 common peaks calibrated on the ground of the Crepis crocea.The similarity and similarity of each batch were above 0.977.A total of 5 common peaks were calibrated at the root;the similarity between the batches of medicinal materials was low between 0.035 and 0.982.3 study on pharmacodynamics of different medical parts of Crepis crocea3.1 Comparison of Antitussive Effects of Different Medicinal Parts of Crepis croceaCompared with the blank group,the cough latency of the model group was significantly prolonged,the number of coughs was significantly increased,and the contents of immune factors IL-4 and IgE in BALF were significantly increased(P <0.05).After administration,compared with the model group,the positive control group,Huanyangshen whole grass group,aboveground grouping,and root group had a significant increase in cough latency(P <0.05),and a significant decrease in the number of coughs within 2 minutes(P <0.05);Each part of Crepis crocea can significantly down-regulate the content of IL-4 in the alveolar lavage fluid of ammonia-induced cough mice(P <0.05);the content of IgE in the whole grass and aboveground alveolar lavage fluid decreased significantly(P <0.05).It is suggested that Huanyang Shenq uancao group,aboveground group,and root group all have a certain antitussive effect on the mouse model of ammonia cough,which may be related to the inhibition of IL-4 and IgE production in lung tissue.3.2 Comparison of Anti-inflammatory Effects of Different Medicinal Parts of Crepis croceaCompared with the blank group,the levels of IL-1β and IL-6 in the model group were significantly increased(P <0.01),indicating successful modeling.After administration,compared with the model group,the positive control group,Huanyangshen whole grass group,aboveground grouping,and root group can significantly reduce the content of leukocytes in the blood(P <0.01),and can significantly reduce serum IL-1β The content of IL-6(P <0.01).It is suggested that Huanyang Shenquancao group,aboveground group and root group all have therapeutic effects on endotoxin-induced inflammation.Among them,Crepis crocea has the strongest healing effect,followed by the ground part of Crepis crocea,and the root healing effect is the worst.3.3 Effects of different medicinal parts of Crepis crocea on cyclophosphamide-induced immunocompromised model miceCompared with the blank group,the levels of immune factors TNF-α and IgG in the model group were significantly increased(P <0.05),and the difference was statistically significant(P <0.05).After administration,the positive control group,Huanyangshenquancao group and root group had a significant downregulation effect on TNF-α(P <0.05);the positive control group,Huanyangshenquancao group had a significant downregulation effect on IgG(P <0.05).Conclusion: Crepis crocea plants are distributed in the four representative areas of grass,grassland,shrub and broad-leaved forest in Shanyin County,but their distribution areas are mostly concentrated in the north and south of Shanyin County,and the distribution range is narrow and the reserves are also Low,suggesting that we need to further understand the growth characteristics of Crepis crocea and increase its resource reserves.The whole grass,aerial parts and roots of Crepis crocea all contain chlorogenic acid,but the aerial parts are significantly higher than the roots,and the polysaccharide content is significantly higher than the aerial parts.difference.The content of extracts varies with the growth period of the plant.The above-ground parts and roots show a certain fluctuation.The content of extracts in the above-ground parts changes from low to high,while the content of roots changes from high to low,which is in line with the law of plant growth and development Ginseng collection has certain guiding significance.By comparing the cough,anti-inflammatory and immunomodulatory effects of different parts,it shows that different parts have the same effect,but the effect intensity is different.The results of the comparison of drug efficacy are consistent with the results of the component studies.The above research provides a basis for the quality control and clinical efficacy research of Crepis crocea and provides a scientific basis for the rational use and promotion of Crepis crocea,a specialty medicine in Shanxi.
