Objective: To optimize extraction process purification process study the chemicalconstituents in phenolic acid from Crepis crocea(Lamk.)Babc. Combining withantiasthmatic active, to establishment fingerprints of HD2, to studie it onSpectrum-effect Relations on HD2of Crepis crocea(Lamk.)Babc. To establishfingerprint Spectrum and to futher clarify the material basis and to perfect the qualitystandard Crepis crocea(Lamk.)Babc.Methods: Based on single factor tests, central composite design-response surfacemethodology was adopted to optimize extraction process; using HPD100macroporous adsorption resin to study purification process of Crepiscrocea(Lamk.)Babc; The phenolic acid from Crepis crocea(Lamk.)Babc. wereisolated by vayious chromatographic techniques and structures were elucidated on thebasis of spectral analyses.To establish fingerprints of HD2, then the standars of IgE、IFNγ、IL-4wereexamined by ELISA. To study fingerprints of HD2and antiasthmatic active bybivariate correlation analysis method. To establish fingerprints of Crepiscrocea(Lamk.)Babc. by Fingerprint similarity evaluation system.Result: Fifteen compounds were obtain and the structures of all compounds wereidentified. Ten of them were first obtain from Crepis crocea(Lamk.)Babc. as asEthylp-hydroxyphenyllactate (1), cis-3,4-Dihydroxy-β-ionion (2), Esculetin(4),3,4-Dihydroxybenzaldehyde (5), trans-4-Hydroxycinnamic acid (6), Methyl p-hydroxyphenyllactate(8),8-β-(2-methyl-2-hydroxy-3-oxobutanoyloxy)-glucozaluzanin C(9),8-Epidesacylcynaropicrin glucoside (10), Tectoroside(13),Cosmosiin(15).Three of them were new compounds as EA3-Ⅰ(3), EA3-Ⅱ(11), EA3-Ⅲ(12). Caffeic acid(7), Luteolin7-O-glucoside(14).Based on the relationship between spectral efficiency research ideas, theapplication of HPLC chromatographic analysis the HD2part due to the chemicalcomposition of Crepis crocea(Lamk.)Babc "spectrum". By examination of thevarious indicators, it shows that the analytical method established in this study isable to meet the technical requirements of fingerprint analysis; Combined with theHD2spectrum analysis and pharmacodynamic relationship, the characteristic peaksof1,2,11,13, and14may be relevant with the antiasthmatic active of Crepiscrocea(Lamk.)Babc. by bivariate correlation analysis method.Conclusion: The subject studies the chemical constituents in phenolic acid fromCrepis turczaniowii systematicly and provides a scientific basis for the developmentof this folk medicinal resources and new datas for the further study of this genus.Based on the spectrum activity relationship, of the establishment of thespectrum-effect relationship successfully find6antiasthmatic active ingredient, andprovide a valuable reference for quality control and efficacy of Crepiscrocea(Lamk.)Babc substance basic research.
