Recurrent oral ulcer is the most common ulcerative disease occurs on the oral mucosa.The onset of the disease may severely affect patients' quality of life.Traditional Chinese medicine has certain advantages in the treatment of this disease.To further understand the mechanism of action of traditional Chinese medicine,the rat model of recurrent oral ulcer was established in this study,and its change rule was studied by using Yangyin Qingre Formulae 1.Yangyin Qingre Formulae 1 is an experience prescription used in the Department of Otorhinolaryngology of the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine,and has a clear clinical effect on recurrent oral ulcers.In this study,the therapeutic effect of Yangyin Qingre Formulae 1 on model rats and its mechanism of action were discussed by observing the general status of the model rats and the expression levels of T cell subsets,CD4 +,and CD8 + values in the peripheral serum,so as to provide a scientific basis for its clinical application.?.Literature Research Recurrent oral ulcer,also known as recurrent aphthous stomatitis,is the most common ulcerative disease that occurs on the oral mucosa,which causes pain and limited function in patients and affects their quality of life.Regarding the etiology of recurrent oral ulcers,there are many theories at different research levels,and most of them have been experimentally and clinically supported,which can be used as a reference to a certain extent.The main view is that the disease is related to the impaired mucosal barrier function,oral microbial flora,dietary nutrition status,and immune factors.Various treatments are currently available and the main is to improve symptoms and promote ulcer healing.CD4 +,CD8 +,and T-cell subsets are immune effector cells that play an important role in lymphocytes.First,in cellular immunity,CD4+ plays an induced and synergistic role,while CD8+ plays an inhibitory and toxic role;the changes of the 2 cell subsets are in opposite directions and coordinate with each other.The subsets are opposed and balanced.When immune function meets unbalance,the number and proportion of lymphocytes are out of balance,leading to the occurrence of disease.Clinical studies and animal experiments show that recurrent oral ulcers may be related to the body's immune dysfunction.Recurrent oral ulcers are called "oral aphthae" in traditional Chinese medicine,which are mainly caused by disorders of body fluid,qi and blood,and yin and yang.Heat evil is the main cause of the disease,and nourishing yin and clearing heat is the root of treating oral aphthae.Traditional Chinese medicine with nourishing yin and clearing heat can reduce the levels of inflammatory factors in patients with recurrent oral ulcers,and at the same time can increase the levels of T cell subsets in patients.Yangyin Qingre Formulae 1 is based on the concept of nourishing yin and clearing heat,with clear clinical effects.Therefore,it is of great significance to further explore the effect of Yangyin Qingre Formulae 1 on the expression levels of T cell subsets,CD4 + and CD8+ in peripheral serum of model rats.?.Experimental research Subjects: take 60 clean SD rats,with half male and half female.The rats were fed to200 ± 20 g for convenience;divide them with random number method into 6 groups,10 in each group,with half male and half female: the material group,pre-test group,model group,traditional Chinese medicine group,western medicine group,and normal group.Methods: Rats from the material group were used to prepare rat oral mucosal antigenic emulsion,and all were killed after the material was collected.The antigenic emulsion of the oral mucosa of rats was prepared by referring to the previous literature,and the rats in the pre-test group were injected.After the successful modeling was confirmed,the same method was applied to the Chinese medicine group,western medicine group and model group.After each successful modeling was confirmed,intragastric administration was performed as follows: 18.23g/Kg/d of crude drug for the traditional Chinese medicine group;vitamin B12 54mg/Kg/d for the western medicine group;the same amount of normal saline for the model group,and no intervention in the normal group,be sure that the time of intragastric administration and modeling is the same in each group.Changes in general state of rats and changes in body weight and water consumption before and after treatment were recorded.The expression of serum T cell subsets and CD4 + / CD8 + values in each group of rats after the experiment were measured by using flow cytometry.Statistical analysis was performed using SPSS 24.0.Results(1)No statistical difference in body weight of rats in each group before the experiment was observed(P> 0.05).After the experiment,the body weight of each group of rats was lowered,and there was a statistical difference compared with the normal group(P<0.05),indicating that oral ulcers in model rats affected food intake.The histogram showed that the model group had the most significant weight loss.Compared with the model group,the rats in the traditional Chinese medicine group and the western medicine group lost slightly less weight(P <0.05),indicating that the treatment was effective.There was no difference in body weight between the Chinese medicine group and the western medicine group(P> 0.05),indicating that the role of Chinese medicine in relieving symptoms and improving eating was equivalent to that of the western medicine group.(2)Rats in each group were fed in cages.There was no statistical difference in the amount of water consumed by rats in each group before the experiment(P> 0.05).After the experiment,the water consumption by the rats in each group increased,and there was a statistical difference compared with the normal group(P <0.05),indicating that rats in the model group consumed more water due to yin deficiency,and the increase in water consumption of the model group was the most significant.Compared with the model group,the rats both in the traditional Chinese medicine group and the western medicine group had less increase in water consumption(P <0.05),indicating that the treatment was effective.Compared with the western medicine group,the water consumption of the Chinese medicine group was different(P <0.05).The water consumption of the Chinese medicine group was slightly lower,indicating that the effect of Chinese medicine on relieving symptoms in rats was better than that of the western medicine group.
