Objective:The classical Xiebaisan is composed of Mori Cortex,Lycii Cortex,licorice and japonica rice,which has the effects of clearing lung heat and relieving asthma and cough.This article studies the standard decoction of the classic Xiebaisan:Improve the quality standards of fried Mori Cortex,Lycii Cortex and fried licorice pieces.Combining research results with modern technology research methods to determine the preparation process of the decoction is intended to provide a basis for the substance standard of Xiebaisan and the development of the preparation.Methods:According to the corresponding methods under the four general rules of the "Chinese Pharmacopoeia" of the 2015 edition to test the characteristics,thin layer,moisture,ash,extracts and other items of the decoction pieces.The content of total flavonoids in fried Mori Cortex was determined by UV;The HPLC was used to establish the fingerprint of fried Mori Cortex,and the content of Kukoamine B inLycii Cortex Pieces and the content of liquiritin and glycyrrhetic acid in fried licorice were determined.In the preparation technology research,check the ancient records to determine the prescription dose and method.For the preparation process with ambiguous parameters,take clinical value as the guide,and use modern research methods to extract paste rate,extract,liquiritin,glycyrrhizic acid content,and fingerprints.The map and other indicators were used to examine the particle size,decoction,filtration,concentration and drying of the decoction pieces,and determine the best preparation process.Results:The characteristics of three decoction pieces were determined.In thin layer chromatography,the reference substance and the test article showed the same fluorescent spots at the corresponding positions,which can be used for identification of decoction pieces.The quality standard of fried Mori Cortex has been preliminarily formulated: the moisture content should not exceed 5.0%,the extract should not be less than 7.0%,the total flavonoids should not be less than 0.4%,and 13 common peaks were identified in the fingerprint.Quality standard of Digupi decoction pieces: water content should not exceed8.0%,extract should not be less than 11.0%,total ash content should not be less than10.0%,acid insoluble ash content should not be less than 3.0%,the content of diospermium B based on dry product should not be less than 1.40%.Quality standard of fried licorice: moisture should not exceed 7.0%,ash should not exceed 5.0%,acid-insoluble ash should not exceed 1.0%,extract should not be less than 30.0%,liquiritin should be no less than 0.45%,and glycyrrhizic acid should be a lot 1.80%.The preparation process of Xiebaisan is: fried Mori Cortex 41.3g,Lycii Cortex pieces 41.3g,fried licorice 4.13 g,three pieces crushed through a 10-mesh sieve,add 10.5g japonica rice to a 3L casserole,add water 1260 mL,use electricity Heating method: decoction under fire until boiling,simmer for 10 minutes(approximately to 882mL),filter with 200 mesh filter cloth while hot,Freeze drying for 60h(T<-40?,Vac<20Pa),then get diarrhea powder Dry paste powder.Conclusion:the quality standards of the decoction pieces have been raised,and the quality of the decoction pieces used in medicine has been controlled from the source.According to the traditional Chinese medicine theory and the textual research based on ancient records,the dosage of Xiebaisan was clarified,and combined with modern research techniques to determine the optimal preparation process of Xiebaisan,which was the benchmark for the following classic xiebaisan Research has laid a solid foundation.
