| ObjectiveTo explore the rules of clinical medicine for treating lung cancer in the Department of Pulmonary Diseases of the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine,and to explore the effects of effective Qi-Yu Sanlong decoction on the growth of subcutaneous transplanted tumor in lung cancer mice,the micromor-Phological changes of tumor cells,and the expression of key autophagy molecule yeast Atg6 homology?Beclin1?,autophag-related gene 5?Atg5?and microtubule-related protein 1 light chain 3?LC3B?.Methods1.The prescription rules of traditional Chinese medicine for lung cancer were discussed based on 792 inpatient records.Data mining technology was used to retrospectively analyze the medical records of 792patients with lung cancer admitted to the first affiliated hospital of anhui university of traditional Chinese medicine from January 2013 to December 2018.The complex network technology was used to construct the core prescription network diagram,SPSS22.0 software was used to calculate the frequency of core TCM medication and the flavor and meridian tropism of prescription drugs.and conduct systematic clustering analysis,then the Aprior module of SPSS Clementine Cliene11.1 software was used to conduct association rule analysis of TCM and association rule analysis of TCM and tumor markers'improvement.2.Effect of Qiyu Sanlong Decoction in Inhibiting Growth of Lung Cancer by Regulating Expressions of Beclin1,Atg5 and LC3BLung cancer mice transplanted model was constructed by inoculation of Lewis lung cancer cells?LLC?into the right forelimb of mice.After successful modeling,the mice was randomly divided into model group,Qi-Yu Sanlong decoction group,chemotherapy group and combined group,with 18 mice in each group.model group?0.2ml/10g Nacl solution by gavage qd+intraperitoneal injection of Nacl 0.4ml at the 1st,3rd and 5th day?;Qi-Yu Sanlong decoction group(80.48 g·kg-1traditional Chinese medicine solution by gavage qd+intraperitoneal injection of Nacl 0.4ml at the 1st,3rd and 5th day);Combined group(80.48g·kg-1traditional Chinese medicine solution by gavage qd+intraperitoneal injection of cisplatin solution?DDP?0.4ml at the 1st,3rd and 5th day);Chemotherapy group?0.2ml/10g Nacl by gavage qd+intraperitoneal injection of cisplatin solution?DDP?0.4ml at the 1st,3rd and 5th day?.The tumor tissue was exfoliated after 21 days of treatment.The tumor weight was measured and the tumor inhibition rate was calculated.The microstructure of tumor cells was observed by Electron microscopy.Immunohistochemistry and Western blot were used to detect the expressions of Beclin1,autophagy-related gene 5?Atg5?,microtubule-related protein 1light chain 3-I?LC3B-I?and microtubule-related protein 1 light chain 3-II?LC3B-II?in tumor tissues,and LC3B-II/LC3B-I was calculated.The transcription levels of Beclin1m RNA and Atg5 m RNA were detected by real-time PCR.Results1.The prescription rules of traditional Chinese medicine for lung cancer were discussed based on 792 inpatient records.According to the analysis of traditional Chinese medicine prescription,the top 20 core drugs were white flower snake tongue herb,poria cocos,half branch lotus,processed licorice root,atractylodes,coicis seed,thunbergii fritillaria,bitter almond,tangerine peel,yellow essence,etc.According to the statistical analysis of the frequency of prescription drug taste normalization,the choice of medicinal property was mainly cold?slightly cold,cool?and warm?slightly warm?,while the choice of medicine taste was mainly bitter?slightly bitter?and sweet?mild?,which mainly went to the lung,spleen and stomach meridian.Core of traditional Chinese medicine association rules analysis shows in the efficacy of the top 10 associated respectively heat medicine to tonify deficiency,phlegm fights medicine to tonify deficiency medicine,tonify deficiency,heat,phlegm fights medicine to clear heat,etc,in the top 10 drugs associated respectively in these-spreading hedyotis herb,spreading hedyotis herb to these fields,poria cocos,atractylodes,spreading hedyotis herb,coix seed,etc.By cluster analysis,six clustering formulae were obtained,including hedyotis partridge-banzhi-huangjing,coix seed-radix pseudostellariae,orange peel-pinellia ginger.The association rules between tumor markers and core drugs showed that the compatibility with panicularia chinensis had the highest correlation with CA153 and SCC-Ag,while the compatibility with astragalus chinensis and astragalus chinensis had the highest correlation with CA125 and CA199.The highest correlation between semen desileiae and TSGF,NSE and CYFRA21-1 was found between semen desileiae and semen desileiae.Zhejiang fritillary bulb compatibility coix seed has the highest correlation degree with CEA.2.Effect of Qi-Yu Sanlong Decoction in Inhibiting Growth of Lung Cancer by Regulating Expressions of Beclin1,Atg5 and LC3BQi-Yu Sanlong Decoction could inhibit the growth of transplanted tumor,with a tumor inhibition rate of 31.2%.Electron microscopy revealed that there were many autophagic vesicles and their contents in tumor cells in Qi-Yu Sanlong Decoction group.Immunohistochemical results showed that Beclin1 and LC3B were mainly distributed in the cytoplasm.Western blot experiments further confirmed that Qi-Yu Sanlong Decoction could up-regulate the expression of Beclin1,Atg5 and LC3B proteins?P<0.01?,and promote the conversion of LC3B-I to LC3-II,compared with the model group.Real-time PCR results showed that Qi-Yu Sanlong Decoction could also promote Beclin1m RNA and Atg5m RNA transcription compared with the model group?P<0.01?.Conclusion1.The clinical treatment of lung cancer is extensive,It is always treated by Tonifying Qi and nourishing yin,invigorating spleen and lung,clearing away heat and detoxification,resolving phlegm and dispersing stagnation,supplemented by products of promoting qi and activating blood circulation,and the strength of strengthening the body and eliminating pathogens should be adjusted according to individual conditions in the clinical treatment to achieve both specimen and treatment.The association rules showed that the compatibility of Hedyotis diffusa,Scutellaria barbata and other anti-pathogenic drugs was associated with the improvement of tumor markers2.Qi-Yu Sanlong Decoction inhibits the growth of transplanted tumor in mice by inducing autophagy in lung cancer cells,and its mechanism may be related to up-regulating the expression of autophagy key molecules Beclin1,Atg5,LC3B and promoting the conversion of LC3B-I to LC3B-II. |
