Objective: To investigate the expression of long non-coding RNA(lncRNA)LINC00473 in hepatocellular carcinoma(HCC)tissues and its relationship with clinicopathological characteristics,to verify the proliferation of LINC00473 in hepatoma cells,The role of migration,invasion and apoptosis has laid a foundation for exploring the molecular mechanism of LINC00473 in the occurrence and development of HCC.Methods: The TCGA database was used to select LINC00473,which was not reported in HCC and was highly expressed.Real-time Quantitative Polymerase Chain Reaction(qRT-PCR)was used to detect the expression level of LINC00473 in 20 liver cancer patients,and the expression level of LINC00473 and the gender,age and TNM stage of the patient were analyzed.Correlation of clinical pathological parameters.The expression levels of LINC00473 in normal hepatocyte Lo2 and HepG2?Huh-7?SMMC-7721?Bel-7402?HCC-LM3?HCC-LM9 were detected by qRT-PCR.Functional experiment.The recombinant plasmid that silenced LINC00473 was transfected into hepatocellular carcinoma cells,and the proliferation was tested at 24-hour and48-hour through 5-Ethynyl-2'-deoxyuridine(EDU)cell proliferation experiment;Cell migration and invasion were detected by Transwell chamber and apoptosis was detected by flow cytometry.Results: It was found in the TCGA database that the expression level of LINC00473 in cancer tissues of liver cancer patients was significantly higher than that of adjacent tissues.In local tissue samples,LINC00473 expression levels were significantly upregulated in HCC cancer tissues relative to paracancerous tissues in local tissue samples(P <0.05).LINC00473 expression levels were the same aspatient age(P<0.05),TNM stage(P<0.05),AJCC clinical stage(P<0.05),AFP(P= 0.001),but was not significantly related to the patient's gender,tumor size,and hepatitis history(P> 0.05).The expression level of LINC00473 in Lo2 of normal liver cells was significantly lower than that of liver cancer cells(P < 0.05).LINC00473 has the highest expression in HepG2.Therefore,this study used HepG2 cells to study the effect of silencing LINC00473 on the function of HCC cells.The experimental results show that silencing LINC00473 can inhibit the proliferation of HepG2 cells;inhibit the cell invasion ability;and effectively promote the apoptosis of HepG2 cells.Conclusion: LINC00473 is up-regulated in HCC cancer tissues and liver cancer cells.Inhibition of LINC00473 can inhibit the proliferation and invasion of HCC cells and promote apoptosis.Initially revealed the role of LINC00473 in the development of HCC.
