ObjectiveTo investigate the effect of protein Z in the progression of atherosclerotic rodents rat model after cerebral ischemia stroke.MethodsRats were randomly divided into the control group and model group.The model group were used to established the atherosclerotic rats model,and the atherosclerotic rats model were randomly divided into five subgroups: the AS-Sham group,the AS rodent model sustained ischemia(AS-SI)1h group,AS-SI 6h group,AS-SI 12 h group and AS-SI 24 h group.We use HE staining to test the pathological changes in the aortic root and the rat brain tissues,and use the Reverse Transcription Polymerase Chain Reactions(RT-PCR)method to analyze the level of protein Z mRNA and the protein Z protein in the tissue were tested by immunohistochemical method.We use the SPSS 16.0 to analyzed the datas and the results were given as the mean ± x.Statistical analysis was performed by ANOVA,P<0.05.Results1.General observation: in edema area,the brain tissues were taken to difference as the time points,and then atrophy and liquefaction necrosis.2.HE Staining: ischemia were taken to neuron displayed in atrophy,necrosis or apoptosis with the time points.3.RT-PCR and IHC: The protein Z expression were significantly up regulated with control group or compare with four-time-points model groups(P<0.05).Protein Z mRNA and the protein productions had changed regularly and reached the peak in the AS-SI 6h group compared with the four-time points.The protein Z mRNA and protein expression are up-regulated with the time progress,in AS-SI 1h group and AS-SI 6h group they were highest expression,in AS-SI 12 group and 24 h group they became lower,but they all above the baseline.In the model group,protein Z expressed widely around the brain micro vascular area,and in the bilateral cerebral cortex and the cytoplasm,protein Z have little around the nucleus.Conclusion1.A stable atherosclerotic model could successfully replicate with the solid-state high fat diet,and then to built for CIS model.2.protein Z expression is synergistic on the regulation in both of tow diseases,in that area the protein Z expressed much higher and stronger.3.There is a closedly relationship between protein Z expression and systematic inflammation.We could built a theoretical model to evaluate the occurrence the cerebral ischemia stroke and study the development or treatment by atherosclerotic disease.
