ObjectiveHypertension by abdominal aorta stenosis in rats and to observe prorenin receptor((Pro)renin receptor(P)RR)expression in rat cardiac muscle protein and its relationship with myocardial remodeling,further explore the mechanism of action of(P)RR in abdominal aortic stenosis in hypertensive rat myocardial reconstruction in the offer the theoretical basis for the prevention and treatment of hypertension.MethodA total of 40 relatively stable blood pressure and quality of the same male SD rats were randomly divided into four groups(10 / group): 1.Sham operation group(Control,C): the removal of ligation of abdominal aorta,the remaining part given the same treatment as hypertension group,normal diet after operation;the high blood pressure.Group(Aortic ligation,AL): hypertensive rat model by abdominal aortic constriction system,normal diet after operation;group.HRP(H): after abdominal aorta constriction(hypertension group)received RR(P)inhibitor HRP(4 g/kg/d)(Ig);4.Group.U73122(U): after abdominal aorta constriction(hypertension group)treated with PLC-beta 3 inhibitor U73122(40 g/kg/d)(Ig);after 4 weeks of testing.Change in the use of noninvasive tail manometry detection of rat blood pressure and myocardial remodeling index;the expression of collagen fibers in myocardial tissue were detected by Masson staining method;using the detection of myocardial Western blot(P)RR,Collagen I,Collagen III,PLC-beta 3 and metalloproteinase MMP2 protein.Result1.Changes of blood pressure,collagen fiber and(P)RR protein expression in rats with abdominal aortic constrictionCompared with the control group,abdominal aortic blood pressure myocardial tissue of hypertensive rats increased significantly in narrow,suggesting that a successful model;collagen fiber in myocardial tissue and the expression level of(P)RR protein increased significantly(P<0.01).2.Effects of(P)RR inhibitors on blood pressure and myocardial remodeling index in hypertensive rats with abdominal aortic stenosisCompared with the sham operation group(107±3mmHg),the arterial blood pressure and myocardial remodeling index in the hypertensive group were significantly increased(P<0.05).After the administration of the HRP inhibitor,compared with the hypertensive group(165±5mmHg),the blood pressure and myocardial remodeling index of group HRP(131±6mmHg)were significantly lower than that of the hypertensive group(P<0.05).3.Effects of(P)RR inhibitors on the expression of collagen fibers in the myocardium of rats with abdominal aortic constriction hypertensionWith the help of Masson staining method showed that abdominal aortic collagen content in myocardial tissue of hypertensive rats was significantly increased compared with that in the narrow control group;HRP inhibitor significantly reduced the expression of the collagen fibers in myocardium.At the same time,the expression of Collagen I and Collagen III protein in myocardium of abdominal aortic constriction hypertensive rats was significantly higher than that in the sham operated group(P<0.05)by Western blot detection.The HRP inhibitor significantly reduced the expression of Collagen I and Collagen III protein in myocardium of abdominal aortic constriction hypertensive rats(P<0.05).4.Effects of(P)RR inhibitors on the expression of PLC-beta 3 and metalloproteinase MMP-2 protein in the myocardium of rats with abdominal aortic constriction hypertensionCompared with the control group,the expression levels of PLC-beta 3 and MMP-2 in cardiac tissue of abdominal aortic constriction hypertensive rats were significantly increased(P<0.05).HRP inhibitors significantly decreased the protein expression of PLC-beta 3 and MMP-2 in rat myocardium(P <0.05).5.Effect of PLC-beta 3 inhibitors on the expression of collagen fiber and metallo-proteinase MMP2 protein in the myocardium of rats with abdominal aorticconstric-tion hypertensionWith the help of Masson staining method showed that abdominal aortic collagen content in myocardial tissue of hypertensive rats was significantly increased compared with that in the narrow control group;PLC-beta 3 inhibitors significantly reduced the expression of the collagen fibers in myocardium;compared with the control group,abdominal aorta myocardial tissue of hypertensive rats in narrow Collagen I and Collagen III in the protein expression level increased significantly(P<0.05).PLC-beta 3 inhibitors significantly reduced the myocardial tissue in Collagen I and Collagen III protein expression(P<0.05);and the detection showed that the expression of protein kinase MMP-2 protein also decreased significantly.Conclusion1.High expression of(P)RR may contribute to myocardial remodeling in hypertensive rats with abdominal aortic constriction;2.(P)RR may participate in the pathological process of myocardial remodeling in rats with abdominal aortic constriction hypertension by means of PLC-beta 3-MMP-2 signaling pathway.
