Application Of KaryoLite^TM BoBs^TM Technology For The Chromosome Evaluation Of Products Of Conceptionfrom From Recurrent Spontaneous Abortion

Purpose Clinically recognized pregnancy loss occurs in approximately 15%-25% of pregnan-cies,with most occurring in the first trimester.About 1%-3% will become to recurrent spontaneous abortion?RSA?.In recent years,more and more people pay attention to the spontaneous abortion,especially the RSA.There are a lot of reasons lead to spontaneous abortion,such as genetic factors,immune factors,infection factors,male specific factors and female specific factors and unknown factors.However,embryo chromosome abnorm-ality is the main cause of early abortion.For the RSA,most detection and treatment are for patients.The detection of abortion itself is very less.If the products of conception were not detected,most causes of misscarrage are unknown.Spontaneous abortion,es-pecially RSA,not only to women's physical damage,but also to the psychological pressure on the family,what is more,the harmony of family will be impacted.This research aims to use KaryoLite^TM BoBs^TM technology to detect the DNA of RSA,so that we can find the chromosomal abnormalities of the fetus in the local area,and make advices for their next gestation.Method KaryoLite^TM BoBs^TM technology,which is baced on bacterial artificial chromosomes?BACs??on?Beads,is a molecular karyotype analysis method.It covers 1-22,X and Y chro-mosome long arm and short arms.We used KaryoLite^TM BoBs^TM technology?PerkinElm-er,Wallac Oy,Turku,Finland?to analyze 25 samples from RSA and 16 samples from spontaneous abortion.All samplems were accepted from chorionic villi,which were come from complete curettage of uterine cavity.All The products of conception were in 6-12 ges-tational weeks.All patients excluded medical disease,and they were nature conceived.Results RSA successfully analysised 25 samples,among which there wre 13 cases of ab-normal chromosome,the abnormal detection rate was 0.52?13/25?.The chromosomal abnormalities were numerical 76.9%?10/13?of which trisomy was the most.Sporadic spontaneous abortion one times chorionic karyotype analysised results of 16 cases,among whicn there were 7 cases of abnormal chromosome.The abnormal detection rate was for43.8%?7/16?,in which abnormal chromosome number was 28.6%?2/7?.There was no significant difference in the incidence of chromosomal abnormalities in spontaneous ab-ortion and RSA.Conclusion: The chromosome abnormal probability between RSA and Sporadic spontaneous abortion was no significant difference.But embryo chromosomal abnormality was the main cause of RSA.Because of the samples were less,it was not clear that the relationship between the abnormal fetal chromosomal abnormality and RSA.Further studies should be made.The KaryoLite^TM BoBs^TM technology for detection of products of conception of RSA had its meanings for the further understanding of the relationship between RAS and chromosome abnormality,so we concluded that it had certain clinical values.Meanings Using the KaryoLite^TM BoBs^TM technology to detect products of conception of RSA,we can analysis of the further reasons.At present,the research like this was less,no matter in domestic or foreign.This study intends to take on domestic and international good experience to explore the clinical value of rapid detection of abortion chromosome through the bacterial artificial chromosomes?BACs??on?Beads.At the same time,we can use the KaryoLite^TM BoBs^TM technology to find the incidence and the distribution condition of chromosomal abnormalities in products of conception of RSA,so that we can give them advices,such as assisted reproduction.
