Objective:The intestinal butyric acid,deoxycholic acid and endotoxin are metabolites produced by intestinal bacteria.Caenorhabditis elegans is a model organism with clear genetic background and simple structure.The dose and time effects of butyric acid,deoxycholic acid and endotoxin on the survival,growth and development of Caenorhabditis elegans were observedfor further understanding their functions.Methods:The subjects were divided into two groupsincluding cgntrol and experimental group.The experimental group was divided into three groupsincluding low,middle and high concentration group.The effects of different concentrations of butyric acid,deoxycholic acid and endotoxin on the growth and anti-infection ability of Caenorhabditis elegans were observed after culturing 48 hours in the experimental group.The movement ability of Caenorhabditis elegans was observed in DO stage(young adult),D4 stage(4th day of adult),D6 stage(6th day of adult).Under the sustained action of butyric acid,deoxycholic acid and endotoxin,the effects of different concentrations of butyric acid,deoxycholic acid and endotoxin on the lifespan and reproductive ability of Caenorhabditis elegans were observed.Results:?The effects of different concentrations of butyric acid on the growth of Caenorhabditis elegans were different,and high concentration of butyric acid could kill Caenorhabditis elegans.Under the sustained action ofdifferentconcentration of butyric acid,the Caenorhabditis elegans' movement ability was enhanced,the life span can be prolonged by butyric acid,but Daf-16::GFP is not localized.Butyric acid can prolong the life span,and reduce the progeny and shorten the spawning time of Caenorhabditis elegans.Different concentrations of butyric acid had no obvious effect on the anti-infection ability of Caenorhabditis elegans.?Deoxycholic acid does not affect the growth of Caenorhabditis elegans.With the increase of concentration and the prolongation of actuation duration,the movement ability of Caenorhabditis elegans was decreased.Middle and high concentration of deoxycholic acid can shorten the life span,reduce ability of the movement and anti-infection,and reduce the progeny and shorten the spawning time of Caenorhabditis elegans.?The effects of different concentrations of endotoxin on the growth and development of Caenorhabditis elegans were different.With the increase of concentration and the prolongation of actuation duration,the ability of movement of Caenorhabditis elegans was decreased,but the frequency of Caenorhabditis elegans pumping decreased slightly,the frequency of Caenorhabditis elegans thrash was the fastest in the D4 stage of Caenorhabditis elegans.Low concentration of endotoxin enhanced the anti-infective ability of Caenorhabditis elegans,while high concentration of endotoxin decreased the anti-infective ability of Caenorhabditis elegans.Under high concentration of endotoxin,the life span can be shorten.There was no difference in the reproductive ability of Caenorhabditis elegans with different concentrations of endotoxin,however,the progeny of the low concentration group was more than the other groups at the end of reproductive.Conclusion:The dose and time effects of butyric acid,deoxycholic acid and endotoxin on the survival,growth and development of Caenorhabditis elegans were different.?Butyric acid does affect the growth of Caenorhabditis elegans.A certain concentration of butyric acid can prolong the life span of Caenorhabditis elegans,reduce the progeny of Caenorhabditis elegans,shorten the spawning time of Caenorhabditis elegans and enhance the movement ability of Caenorhabditis elegans.High concentration of butyric acid is lethal to Caenorhabditis elegans.Butyric acid prolonged the lifespan of Caenorhabditis elegans,but Daf-16::GFP is not localized.Butyric acid has no influence on the anti-infection ability of Caenorhabditis elegans.?Deoxycholic acid does not affect the growth of Caenorhabditis elegans.High concentration of deoxycholic acid have chronic toxicity and reproductive toxicity,the mechanism of shortening the life span,reducing ability of the movement and anti-infection should be further studied.?Endotoxin does affect the growth and development of Caenorhabditis elegans.Under high concentration of endotoxin,the life span of Caenorhabditis elegans was shortened and the ability of movement was decreased.The anti-infective ability of Caenorhabditis elegans was enhanced by low concentration of endotoxin.Endotoxin has no influence on the reproduction of Caenorhabditis elegans.The anti-infection ability of butyric acid and the reproductive effect of endotoxin on Caenorhabditis elegans need to be further studied.
