| [Objective]To obtain the genotype and allele frequency distribution of DDAH1 gene rs233113 polymorphism in type 2 diabetes mellitus in Yunnan,and to investigate DDAH1 gene rs233113 polymorphism and plasma ADMA level,NO level and type 2 correlation of diabetic atherosclerosis.[Methods]Using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)to treat 100 healthy controls(NGT)and 200 patients with type 2 diabetes.Polymorphism of DDAH1 gene rs233113 in 72 patients with type 2 diabetes mellitus(T2DM)and 128 patients with type 2 diabetes mellitus complicated with atherosclerosis(T2DM-AS);detection of plasma ADMA by enzyme-linked immunosorbent assay(ELISA)The concentration of plasma was determined by nitrate reductase method,and carotid IMT was detected by color Doppler ultrasound and compared with clinical indicators.The distribution of genotypes and alleles in the three groups was tested by 2 tests;the normal distribution data were expressed as mean±standard deviation(x±s).The two groups were compared by independent sample t test,and the comparison between groups was analyzed by one-way ANOVA.(ANOVA),the variance is compared with LSD-t for pairwise comparison,the variance is used by Tamhane,s method for pairwise comparison;the non-normal distribution data is expressed by median±(P25%,P75%).The Mann-Whiney test was used for comparison between the two groups.The Kruskal-Wallis test was used for comparison between the three groups.The correlation was analyzed by Spearman rank correlation analysis.Logistic regression analysis was used for risk factor analysis.[Results]①Plasma ADMA levels in the three groups,T2DM-AS group(1.24±0.43 umol/L)>T2DM group(1.06±0.36 umol/L)>NGT group(0.60±0.32 umol/L),(P<0.05);②plasma NO levels in the three groups:NGT group(76.90±15.08 umol/L)>T2DM group(62.60±10.06 umol/L)>T2DM-AS group(58.34±9.43 umol/L),(P<0.05);③DDAH1 gene rs233113 A/A,A/T+T/T genotype frequency was 49.2%,50.8%in T2DM-AS group,44.4%,55.6%in T2DM group,and NGT group 41%,59%;the difference between the three groups was not statistically significant(χ2=1.713,p=0.425);④the two alleles of the DDAH1 gene rs233113 locus A and T frequencies in type 2 diabetes with atherosclerosis 70.1%,29.9%;70.8%,29.2%in the type 2 diabetes group,66%,34%in the healthy control group,respectively;the difference between the three groups was not statistically significant(χ2=1.201,p=0.549);⑤spearson correlation analysis showed that plasma ADMA levels in T2DM-AS group were positively correlated with age,postprandial insulin,carotid IMTand HOMA-IR(r=0.045,0.003,0.001,0.047,P<0.05);⑥T2DM-AS plasma No correlation was found between NO concentration and other indicators.⑦T2DM-AS group carrying AA genotype plasma NO was higher than carrying AT+TT genotype,60.06±9.06,56.68±9.56,P<0.05;carrying AA genotype individuals plasma ADMA level,carotid IMT ratio carrying AT+TT genotype were low,1.07±0.35,1.40±0.41,1.09±0.17,1.21±0.29,(P<0.05);⑧Logistic regression showed that plasma ADMA levels,age,fasting blood glucose,systolic blood pressure may be independent risk factors for atherosclerosis in type 2 diabetes(P=0.021,0.001,0.001,0.025,OR=3.926,1.103,2.272,1.043);plasma NO levels and high-density lipoprotein may be protective factors for atherosclerosis in type 2 diabetes(P=0.018,0.013,OR=0.951,0.064);⑨plasma ADMA ROC curve area is 0.610;[Conclusions]①The DDAH1 gene rs233113 polymorphism exists in the Han population in Yunnan.②This study has not found the DDAH1 gene rs233113 polymorphism and type 2 diabetes atherosclerosis in the Han population of Yunnan.Correlation between;③plasma ADMA levels,age,fasting blood glucose,systolic blood pressure may be an independent risk factor for atherosclerosis in type 2 diabetes;④ plasma NO levels,high-density lipoprotein may be a protective factor for the development of atherosclerosis in type 2 diabetes,and DDAH1 gene rs233113 polymorphism may be unrelated to type 2 diabetes atherosclerosis;⑤The ROC curve suggests that plasma ADMA level has a predictive value for atherosclerosis in type 2 diabetes. |
