| Objective:The aim of this study was to investigate the association of growth arrest specific protein 6(GAS6)gene rs8191974 polymorphism and plasma level with type 2 diabetes mellitus in Kunming Han population.Methods:Peripheral venous blood was collected from 352 patients with type 2 diabetes and 186 normal controls.Then plasma and blood cells were separated.Whole blood genomic DNA extraction kit was used to extract blood cell DNA.Then the PCR was carried out.The polymorphism of the PCR products was detected by one-way gene sequencing method.The plasma GAS6 level was detected by ELISA,and the genotype frequency,allele frequency,plasma GAS6 level and related clinical indexes of T2DM and NC group were compared and analyzed.Results:(1)The gene frequency of GAS6 gene rs8191974 in subjects of T2DM and NC group were consistent with the law of Hardy-Weinberg genetic equilibrium(P>0.05)(2)The frequency of GG genotype and G allele in T2DM group were higher than those in NC group(x2=11.360,P=0.003;x2=11.337,P=0.001).(3)Compared with NC group,the levels of BMI,SBP,DBP,TC,TG,LDL and FBG were higher,but HDL,Cr and GAS6 were lower in T2DM group(P<0.05).(4)The SBP level of GG genotype carriers was higher than that of AA genotype carriers,and the level of DBP was higher than that of GA genotype carrers,while the level of HDL and GAS6 were lower than that of GA and AA genotype carriers(P<0.05).(5)Spearman correlation analysis showed that there were negative correlation between plasma GAS6 level and DBP(r=-0.135,P<0.05),HbAlc(r=-0.210,P<0.05),FBG(r=-0.322,P<0.05)and HOMA-IR(r=-0.215,P<0.05),and positive correlation with age(r=0.153,P<0.05).After controlling age and gender for partial correlation analysis,the results showed that there was no correlation between plasma GAS6 level and DBP,but there were still negative correlation between plasma GAS6 level and HbAlc(r=-0.217,P<0.05),FBG(r=-0.344,P<0.05)and HOMA-IR(r=-0.186,P<0.05).(6)Logistic regression analysis showed that plasma GAS6(OR=0.981;95%CI:0.964-0.998)and GA and AA genotype of GAS6 gene rs8191974(OR value:0.653,0.434,respectively;95%Cl:0.432-0.987,0.190-0.991,respectively)may be the protective factors for T2DM.BMI and TG are the risk factors for T2DM(OR value:1.203,1.332,respectively;95%CI:1.120-1.293,1.113-1.593,respectively).Conclusions:(1)The risk of T2DM in carriers with GAS6 rs8191974 GG genotype was higher than that in carriers with GA and AA genotype.(2)The level of plasma GAS6 may be negatively correlated with the occurrence of T2DM in Kunming Han population.(3)High BMI and triglyceride are risk factors of T2DM-Objective:The aim of this study was to investigate the association of GAS6 gene polymorphism and plasma level with type 2 diabetes mellitus complicated with atherosclerosis in Kunming Han population.Methods:Peripheral venous blood was collected from 186 normal controls,181 patients with type 2 diabetes and 171 patients with type 2 diabetes complicated with atherosclerosis.Then plasma and blood cells were separated.Whole blood genomic DNA extraction kit was used to extract blood cell DNA.Then the PCR was carried out.The polymorphism of the PCR products was detected by one-way gene sequencing method.The plasma GAS6 level was detected by ELISA,and the genotype frequency,allele frequency,plasma GAS6 level and related clinical indexes of NC,T2DM and T2DM-AS group were compared and analyzed.Results:(1)There was no difference in the frequency distribution of the three genotypes between T2DM group and NC group(χ 2=8.031,P=0.018).Compared with NC group,the frequency of GA and AA genotype was lower,but GG genotype was higher in T2DM-AS group(χ2=9.286,P= 0.010).(2)Compared with NC group,the frequency of allele A was lower,but allele G was higher in T2DM and T2DM-AS groups(χ2=7.493,P=0.006;χ2=8.718,P=0.003).There was no difference in allele frequency between T2DM-AS and T2DM group(χ2=0.069,p=0.793).(3)Compared with NC group,the levels of BMI,SBP,DBP,TC,TG,LDL and FBG were higher(P<0.017),but the levels of HDL and GAS6 were lower in T2DM and T2DM-AS group(P<0.017).Compared with T2DM group,the course of T2DM,SBP,and IMT were higher in T2DM-AS group(P<0.017),but there was no difference in GAS6 level between T2DM and T2DM-AS group(P=0.545).(4)Logistic regression analysis showed that the course of T2DM,alcohol consumption and SBP are the risk factors of atherosclerosis(OR values:1.072,2.625,1.019,respectively;95%CI:1.024-1.122,1.241-5.552,1.003-1.035,respectively).HDL is a protective factor for atherosclerosis(OR values:0.253;95%CI:0.071-0.902).However,GAS6 gene rs8191974 polymorphism and plasma level may not be associated with atherosclerosis.Conclusions:(1)The association of GAS6 gene rs8191974 polymorphism and plasma level with atherosclerosis has not been found in Kunming Han population.(2)The course of T2DM,alcohol consumption and hypertension are risk factors for atherosclerosis.(3)HDL is a protective factor for atherosclerosis. |
