OBJECTIVE From the perspectives of pharmacodynamics and material basis,and to explore the difference in the effect of different blood combinations of angelica sinensis,it provides an experimental basis for the theory " pungent flavor make blood circulate".of compatibility of angelica sinensis.METHODS1.Seventy SD rats were randomly divided into blank group,model group,positive drugpair group,angelica with codonopsis group,angelica with zingiber group,angelica with carthamii Flos group and angelica with cyperus rotundus group.The blank group and the model group were given normal saline,and the administration group was administered with compound Danshen dripping pill solution,angelica with codonopsis decoction,angelica with zingiber decoction,angelica with carthamii Flos decoction,and angelica with cyperus rotundus decoction.The acute blood stasis rat model was replicated by subcutaneous injection of epinephrine hydrochloride and ice water bath,with whole blood viscosity(WBV),plasma viscosity(PV),blood sedimentation equation K value(ESRK),erythrocyte aggregation index(EAI),hematocrit.(HCT),prothrombin time(PT),thrombin time(TT),fibrinogen(FIB),heart and lung tissue pathological changes were used as indicators,and to observed the difference of blood-activating effect of angelica sinensis with different taste drugs.2.Sixty-four SD rats were randomly divided into blank group,model group,positive drugpair group,angelica with aconite group,angelica with zingiber group,angelica with cinnamon group and angelica with evodia rutaecarpa group.The blank group and the model group were given normal saline,and the administration group was administered with compound Danshen dripping pill solution,angelica with aconite decoction,angelica with zingiber decoction,angelica with cinnamon decoction,and angelica with evodia rutaecarpa decoction.The same acute blood stasis rat model was used,with common carotid artery systolic pressure(SAP),common carotid artery diastolic pressure(DAP),difference pressure(DP),heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular diastolic pressure(LVDP),maximum increase rate of left ventricular internal pressure(+dp/dt),maximum decrease rate of left ventricular internal pressure(-dp/dt),whole blood viscosity(WBV),plasma viscosity(PV),blood sedimentation equation K value(ESRK),erythrocyte aggregation index(EAI),hematocrit.(HCT),pathological changes of heart and lung tissues were used as indicators,and to observe the difference of blood-activating effect of Angelica sinensis with different pungent and heat drugs.3.Use the following chromatographic conditions: the chromatographic column was Agilent ZORBAX SB-C18 column(4.6 mm×250 mm,5 ?m);mobile phase was 0.1% aqueous formic acid(A)-acetonitrile(B);gradient elution(0 ~ 50 min,5% ~ 40% B 50-60 min,40%-100% B;60-70 min,100% B);detection wavelength: 280 nm,254 nm;volume flow 0.5 mL/min;injection volume 10,20 ?L;column temperature 25 °C,to establishing the High Performance Liquid Chromatography(HPLC)characteristic map of Angelica with different herbs of warm.RESULTS1.Compared with the model group,the compatibility of angelica with codonopsis group could significantly reduce WBV and EAI(P < 0.05),which significantly improved the phenomenon of myocardial cell widening and pulmonary hemorrhage;angelica with zingiber group can significantly reduce WBV,PV,HCT,ESRK,EAI(P < 0.05,P < 0.01),prolonged PT,TT values(P < 0.05,P < 0.01),the myocardial cell spacing change wide,alveolar wall thickening is better than other combinations;angelica with carthamii Flos group can significantly reduce WBV,PV,HCT and ESRK(P < 0.05,P < 0.01),extended PT(P < 0.01),the nucleus became round,lung machine edema was improved;angelica with cyperus rotundus group could reduce WBV at high shear rate and HCT(P < 0.05),no significant improvement of coagulation function,the myocardial cells widened,pulmonary interstitial edema phenomenon has certain improvement.The improvement of WBV and EAI in the angelica with zingiber group was significantly better than that in the angelica with carthamii Flos group and the angelica with cyperus rotundus group,and the improvement of PV was significantly better than that in the angelica with codonopsis group and the angelica with cyperus rotundus group,and the TT in the angelica with zingiber group was significantly longer than that in the other treatment groups.2.Compared with the model group,the angelica sinensis group significantly increased LVSP(P < 0.05)and decreased DP,WBV,PV,HCT,ESRK and EAI(P < 0.05,P < 0.01);the angelica with aconite group significantly increased LVSP(P < 0.05)and decreased DP,WBV,PV,HCT,and EAI(P < 0.05,P < 0.01);the angelica with zingiber group significantly increased LVSP(P < 0.05)and decreased DP,HR,WBV,PV,HCT,ESRK and EAI(P < 0.05,P < 0.01);the angelica with cinnamon group significantly increased LVSP(P < 0.05)and decreased DP,WBV,PV,HCT and ESRK(P < 0.05,P < 0.01);the angelica with evodia rutaecarpa group significantly increased DAP and LVSP(P < 0.05)and decreased DP,WBV,PV,HCT,ESRK and EAI(P < 0.05,P < 0.01).Every angelica drug pairs on the myocardium,lung lesions were improved.The improvement of HCT in the angelica with evodia rutaecarpa group was significantly better than that in the angelica group,the improvement of EAI was significantly better than that in the angelica with cinnamon group,and the improvement of HR was significantly better than that in other treatment groups.3.Fifteen characteristic peaks(f1-f15)were isolated from angelica with aconite drug pair,the relative peak area of the characteristic components ferulic acid,ligustilide and benzoyl aconitine decreased after decoction;19 characteristic peaks(g1-g19)were isolated from angelica with zingiber drug pair,after decoction,the relative peak areas of ferulic acid,ligustilide and 6-gingerol decreased,and the relative peak area of 8-gingerol increased;16 characteristics peaks(r1-r16)were isolated from angelica with cinnamon drug pair,after decoction,the relative peak areas of ferulic acid and cinnamaldehyde increased,and the relative peak areas of ligustilide and cinnamic acid decreased;23 characteristic peaks(w1-w23)were isolated from angelica with evodia rutaecarpa drug pair,after the decoction,the relative peak areas of ferulic acid,ligustilide and rutaecarpine were all increased,and evodiamine disappeared.Among them,the peak areas of r14,w1-w3,w6-w8,w10,w13,w14,w16,w22,w23 increase;f3,f4,f6,f7,f9,f11,f13,f15,g2,g6,g9,g15-g17,g19,r2,r4,r7,r8,r12,r15,r16,w4,w9,w11,w17,w20 peak area decreased;f1,f2,f10,g4,r13,w5,w18 peak area unchanged,the peaks f8,f12,f14,r6,w12,w15,w19 and w21 disappeared,and the peaks g5,g7 and g13 were added.f6,g9,r9 and w13 are ferulic acid,f15,g19,r16 and w23 are ligustilide,f7 is benzoylmesaconine,f10 is benzoylhypacoitine,g17 is 6-gingerol,g18 is 8-gingerol,r12 is cinnamic acid,r14 is cinnamaldehyde,W21 is evodiamine and w22 is rutecarpine.The change in the content of these characteristic components may be the main cause of the difference in the effect of Angelica durg pairs on blood circulation.CONCLUSION1.Angelica with Codonopsis drug pair,Angelica with Zingiber drug pair,Angelica with Carthamii Flos drug pair and Angelica with Cyperus rotundus drug pair can improve in various degree of acute blood stasis rat blood rheology,coagulation system and the degree of pathological damage,the effect of Angelica with Zingiber drug pair is better than the other 3 groups.2.The combination of angelica with different pungent and heat drugs(aconite,zingiber,cinnamon and evodia rutaecarpa)can improve the hemodynamics,hemorheology and pathological damage of rats with acute blood stasis,the effect of Angelica with evodia rutaecarpa drug pair is better than the other 3 groups.3.The content of characteristic components of Angelica sinensis with herbs of warm has different changes,and not only the individual components disappear,but also the formation of new components.The method of HPLC-established Angelica with different herbs of warm is simple and repeatable.
